Role and Significance of NF-kappa B Signaling Pathway in Regulation of Epithelial-mesenchymal Transition in Invasion and Metastasis of Cervical Cancer

摘要 探讨NF-κB信号通路调控上皮-间质转化在宫颈癌侵袭和转移机制中的作用及意义。收集73例宫颈癌患者作为研究对象,其中宫颈浸润癌患者的病灶组织47例和宫颈癌未发生浸润患者的病灶组织26例,选择NF-κB信号通路的标志蛋白NF-κB及其上游调控因子TNF-α、下游调控因子IL-8为技术指标,采用免疫组织化学方法进行检测。①经免疫组化实验,实验结果表明宫颈癌未发生浸润、浸润癌无淋巴转移、浸润癌有淋巴转移的病灶组织中NF-κB蛋白的表达率分别为23.08%、59.09%、92.00%,TNF-α阳性率分别为11.54%、54.55%、84.00%,IL-8阳性率分别为15.38%、59.09%、96.00%,在发生浸润及淋巴结转移后,NF-κB、TNF-α、IL-8的表达显著高于未发生浸润组织(P<0.05)。②浸润性宫颈癌组织中NF-κB、TNF-α及IL-8表达与组织学分级密切相关。NF-κB信号通路通过调控上皮-间质转化影响了宫颈癌的侵袭转移过程,可以为宫颈癌的靶向治疗提供理论依据。 To investigate the role and significance of NF-kappa B signaling pathway in epithelial-mesenchymal transition(EMT)in invasion and metastasis of cervical cancer,seventy-three cervical cancer patients were collected as the study objects,among whom,47 cases of invasive cervical cancer(observation group)and 26 cases of cervical carcinoma in situ(control group)were collected.The marker protein of NF-kappa B signaling pathway,its upstream regulator TNF-alpha and downstream regulator IL-8 were selected as the technical indicators and detected by immunohistochemical method.(1)The expression rates of NF-kappa B protein were23.08%,59.09%and 92.00%respectively in the lesion tissues without infiltration of cervical cancer,without lymph node metastasis,and has lymph node metastasis.The positive expression rates of TNF-αwere respectively 11.54%、54.55%and 84.00%,the positive expression rates of IL-8 were 15.38%、59.09%and 96.00%respectively.Immunohistochemistry showed that the expression of NF-kappa B,TNF-alpha and IL-8 in 47 cases of cervical cancer was significantly higher than that in situ cancer(P<0.05)after infiltration and lymph node metastasis.(2)The expression of NF-kappa B,TNF-alpha and IL-8 in invasive cervical cancer was closely related to histological grade.NF-kappa B signaling pathway affects the invasion and metastasis of cervical cancer by regulating epithelial-mesenchymal transition,which could provide theoretical basis for targeted therapy of cervical cancer.
