β-榄香烯作用于JAK2/STAT3通路抑制肝癌细胞增殖、迁移和侵袭行为 被引量:2

β-elemene inhibits the proliferation,migration and invasion of hepatocellular carcinoma cells through JAK2/STAT3 pathway

摘要 目的探究β-榄香烯通过Janus蛋白酪氨酸激酶2/信号转录及转录激活因子3(janus protein tyrosine kinase 2/signal transducer and activator of transcription 3,JAK2/STAT3)通路对肝癌细胞增殖、迁移和侵袭行为的作用。方法体外培养人肝癌细胞系HepG2,分为对照组、β-榄香烯低、中、高剂量组(L、M、H组),对照组细胞不做处理,β-榄香烯L、M、H组分别加入β-榄香烯至终浓度10、20及40μg/mL。采用MTT法检测HepG2细胞增殖能力,采用划痕实验检测细胞迁移能力,采用Transwell实验检测细胞侵袭能力,Western blot检测JAK2/STAT3信号通路蛋白表达情况。结果与对照组比较,β-榄香烯-L、M、H组细胞活力降低,侵袭细胞数减少,划痕愈合率降低,p-JAK2/JAK2、p-STAT3/STAT3下调(P<0.05);与β-榄香烯-L组比较,β-榄香烯-M、H组细胞活力降低,侵袭细胞数减少,划痕愈合率降低,p-JAK2/JAK2、p-STAT3/STAT3下调(P<0.05);与β-榄香烯-M组比较,β-榄香烯-H组细胞活力降低,侵袭细胞数减少,划痕愈合率降低,p-JAK2/JAK2、p-STAT3/STAT3蛋白表达下调(P<0.05)。结论β-榄香烯能够抑制肝癌细胞增殖、侵袭及转移等行为及JAK2/STAT3通路活性。 Objective To investigate the effect of β-elemene on proliferation,migration and invasion of hepatocellular carcinoma cells through janus protein tyrosine kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3)pathway.Methods Human hepatocellular carcinoma cell line HepG2 was cultured in vitro and divided into control group,low(L),medium(M)-and high(H)-dose β-elemene groups.Cells in the control group were not treated,and β-elemene final concentrations of 10,20,40μg/mL were added to the L,M and H dose groups,respectively.The proliferation ability of HepG2 cells was detected by MTT assay,the migration ability of HepG2 cells was detected by scratch assay,and the invasion ability of HepG2 cells was detected by Transwell assay.The protein expression of JAK2/STAT3 signaling pathway was detected by Western blot.Results Compared with the control group,the cell viability,invasive cell number,wound healing rate and protein expression of p-JAK2/JAK2 and p-STAT3/STAT3 were decreased in β-elemene-L,M and H dose groups(P<0.05).Compared with the β-elemene-L dose group,the cell viability,the number of invasive cells,the wound healing rate and p-JAK2/JAK2 and p-STAT3/STAT3 were decreased in the β-elemene-M and H dose groups(P<0.05).Compared with β-elemene-M dose group,the cell viability,invasive cell number,scratch healing rate and p-JAK2/JAK2 and p-STAT3/STAT3 in β-elemene-H dose group were significantly decreased(P<0.05).Conclusion β-elemene can inhibit the proliferation,invasion and metastasis of hepatocellular carcinoma cells,and the activity of JAK2/STAT3 pathway.
