狗尾草多酚的提取工艺及抗氧化活性研究 被引量:15

Extraction process and antioxidant activity of polyphenols from Setaria viridis

摘要 以狗尾草为原料,研究狗尾草中多酚的提取工艺和体外抗氧化活性。在单因素试验的基础上,采用响应面法优化狗尾草多酚的超声波辅助提取工艺,以抗氧化剂二丁基羟基甲苯(BHT)264为阳性对照,对其羟基自由基、DPPH自由基、超氧阴离子自由基清除能力进行研究。结果表明,在超声功率250W的条件下,最佳提取工艺为料液比1∶20(g∶mL)、乙醇体积分数70%、提取温度80℃、提取时间75min,此条件下多酚提取率平均值为3.23%,与理论值3.45%相近,最佳工艺具有可行性,提取得到的狗尾草多酚对羟基、超氧阴离子、DPPH自由基的半抑制浓度(IC50)分别为1.217、0.476、0.227,当样品浓度达到试验最大浓度时,清除率分别为(48.56±3.46)%、(88.05±2.95)%、(65.34±4.19)%,表现出较好的体外抗氧化活性。 Using Setaria viridis as raw material,the extraction process and antioxidant activity in vitro of polyphenols from S. viridis were studied. On the basis of single factor experiments,the ultrasonic assisted extraction process of polyphenols was optimized by response surface methodology. Using antioxidant butylated hydroxytoluene (BHT) 264 as a positive control,the scavenging ability of hydroxyl radical,DPPH radical and superoxide anion radical were studied. The results showed that under the condition of ultrasonic power 250 W,the optimal extraction process was solid to liquid ratio 1∶20 (g∶ml),ethanol volume fraction 70%,extraction temperature 80 ℃,and time 75 min. The average extraction rate of polyphenols under this condition was 3.23%,which was similar to the theoretical value of 3.45%,and the optimal process was feasible. The half maximal inhibitory concentration (IC50) of the extracted polyphenols to hydroxyl radical,superoxide anion radical and DPPH radical were 1.217,0.476 and 0.227,respectively. When the sample concentration reached the maximum concentration of the test,the scavenging rates were (48.56±3.46)%,(88.05±2.95)% and (65.34±4.19)%,respectively,which showed in vitro good antioxidant activity.
