miR-142在乳腺癌组织和细胞中的表达变化及其对MDA-MB-231细胞侵袭迁移能力的影响 被引量:4

摘要 目的观察微小RNA-142(miR-142)在乳腺癌组织和细胞中的表达变化及其对乳腺癌细胞侵袭转移的影响。方法收集手术切除的92例乳腺癌组织及相对应癌旁组织,体外培养人乳腺癌细胞MCF-7、MDA-MB-231、MDA-MB-468及人正常乳腺上皮细胞HBL-100,采用qRT-PCR法检测组织和细胞中的miR-142。将MDA-MB-231细胞分为观察组和对照组,分别转染miR-142模拟物及阴性对照序列,采用qRT-PCR法检测细胞中的miR-142,分别采用Transwell小室侵袭及迁移实验观察转染后细胞的侵袭及迁移能力。结果乳腺癌组织中miR-142相对表达量为0. 35±0. 09,低于癌旁组织中的1. 02±0. 04(P <0. 01)。乳腺癌MCF-7、MDA-MB-231、MDA-MB-468细胞中miR-142相对表达量分别为0. 53±0. 05、0. 17±0. 06、0. 19±0. 08,均低于人正常乳腺上皮细胞HBL-100中的1. 01±0. 03(P均<0. 01),且高侵袭性乳腺癌细胞MDA-MB-231、MDA-MB-468中miR-142相对表达量低于低侵袭性乳腺癌细胞MCF-7(P均<0. 01)。转染后观察组MDA-MB-231细胞中miR-142相对表达量为32. 97±1. 89,高于对照组的1. 01±0. 06(P <0. 01)。观察组迁移和侵袭实验中穿膜细胞分别为(110±15)、(52±9)个,均低于对照组的(305±27)、(148±16)个(P均<0. 01)。结论 miR-142在乳腺癌组织和细胞中表达降低,且高侵袭性低于低侵袭性乳腺癌细胞;提高乳腺癌细胞中miR-142的表达水平,能够抑制细胞的侵袭和迁移能力。 Objective To explore the expression of microRNA(miRNA)-142 in the breast cancer tissues and cells,and its effect on invasion and metastasis of breast cancer cells.Methods Ninety-two breast cancer tissues and the corresponding para-carcinoma tissues were collected,and human breast cancer cell lines MCF-7,MDA-MB-231 and MDA-MB-468,as well as the human normal mammary epithelial cell line HBL-100,were cultured in vitro.The miR-142 expression in tissues and cells was detected by using qRT-PCR.MDA-MB-231 cells were divided into the observation group and control group,which were then transfected with miR-142 mimics and negative control sequence,respectively.Furthermore,miR-142 was detected by qRT-PCR,and the invasion and migration of the transfected cells were observed by Transwell invasion and migration assay.Results The relative expression of miR-142 in the breast cancer tissues was 0.35±0.09,which was lower than that(1.02±0.04)in the adjacent tissues(P<0.01).The relative expression levels of miR-142 in the MCF-7,MDA-MB-231 and MDA-MB-468 cells of breast cancer were 0.53±0.05,0.17±0.06,and 0.19±0.08,respectively,which were lower than that(1.01±0.03)in the human normal mammary epithelial cells HBL-100(all P<0.01),and the relative expression levels of miR-142 in the high invasive breast cancer cells MDA-MB-231 and MDA-MB-468 were also lower than those in the low invasive breast cancer MCF-7 cells(all P<0.01).After transfection,the relative expression of miR-142 in the MDA-MB-231 cells of the observation group was 32.97±1.89,which was higher than that in the control group(1.01±0.06)(P<0.01).In the experiment of migration and invasion,the number of penetrating cells in the observation group was 110±15 and 52±9,respectively,which were lower than that of the control group[(305±27)and(148±16)](both P<0.01).Conclusions The expression of miR-142 in the breast cancer tissues and cells decreases,and the expression of miR-142 in highly invasive breast cancer cells is lower than that of low invasive breast cancer cells.Up-regulating miR-142 level in breast cancer cells can inhibit the invasion and migration of cancer cells.
