摘要
肿瘤耐药是由药物靶标改变、细胞内部药物浓度下降等引起的复杂过程。尽管新型化疗药物的设计正在迅速发展,但对于晚期肿瘤(如侵袭和转移)尚无有效的化疗药物。肿瘤细胞对药物的耐药性受多种因素影响,如个体遗传差异、肿瘤微环境、肿瘤干细胞、药物失活、药物吸收减少、药物代谢改变、药物靶点基因突变、肿瘤免疫系统改变等。本文介绍了目前主要的肿瘤耐药发生机制以及肿瘤对常见抗肿瘤药物耐药的原因,并从不同角度总结解决肿瘤耐药性的策略。
Cancer drug resistance is a complex process caused by changes in drug targets and the concentration reduction in the cells and so on.Although the design of new chemotherapeutic drugs is evolving rapidly,effective chemotherapeutic drugs have not been found for advanced cancers such as invasion and metastasis.Resistance of cancer cells to therapeutic agents is influenced by a number of factors,such as genetic differences among different tumor microenvironment,tumor stem cells,drugs inactivation,drug absorption deduction,anti-cancer drug metabolism alteration,target gene mutation and tumor immune system changes.In this review,we highlight the current major cancer drug resistance mechanisms and the failure of common therapy drugs in different types of cancer,and summary the main solutions to the problem.
