大鼠脑缺血再灌注后线粒体ATP酶活性、形态学变化及丹参多酚酸的保护作用 被引量:22

Effects of Salvianolateon on ATPase activity of mitochondria and pathomorphology in rats with cerebral ischemia reperfusion

摘要 目的观察注射用丹参多酚酸对大鼠脑缺血再灌注损伤后线粒体ATP酶活性、及形态学的影响。方法将54只雄性SD大鼠随机分成3组(n=18):假手术组(Sham组)、缺血再灌注组(IR组)、丹参多酚酸组(Salvianolate组)。HE染色法观察大脑神经元的组织病理学变化;TTC染色检测脑梗死体积;分光光度计发法线粒体丙二醛含量和线粒体Na^+/K^+ATP酶、Ca^(2+)ATP酶、Mg^(2+)ATP酶活性。结果假手术组大鼠脑组织红染,未见梗死灶;丹参多酚酸组神经细胞变性坏死程度、脑梗死面积较缺血再灌注组明显减轻;与缺血再灌注组相比,丹参多酚酸组线粒体丙二醛含量下降(P<0.05)和线粒体Na^+/K^+ATP酶、Ca^(2+)ATP酶、Mg^(2+)ATP酶活性升高(P<0.05)。结论注射用丹参多酚酸对缺血再灌注损伤有保护作用,其机制与提高线粒体ATP酶活性有关。 Objective To explore the effect of Salvianolate on ATPase activity induced by focal cerebral ischemia in rats.Methods 54 male Sprague Dawley(SD)rats were randomly divided into 3 groups(n=18):sham group,ischemia-reperfusion group(I/R)and Salvianolate group.Histopathological changes of cortex neurons were observed under HE staining.Infarct volumes were measured by staining with triphenyltetrazolium chloride(TTC),and activities of Na+/K+ATPase,Ca 2+ATPase,Mg 2+ATPase of mitochondria were measured by spectrophotometer.Result The brain’s cortex of sham group showed red,indicating no infarct.Salvianolate group,compared with the ischemia-reperfusion group,significantly reduced Degeneration of necrotic cells,infarct size(P<0.05).The activities of Na+/K+ATPase,Ca 2+ATPase,Mg 2+ATPase of mitochondria in the Salvianolate group,compared with the ischemia-reperfusion group,were noticeably increased(P<0.05),however,the levels of MDA was decreased(P<0.05).Conclusion Salvianolate has a neuroprotective effect in focal cerebral ischemia rats.The mechanism is associated with Increased ATPase activity of mitochondria.
