正交法优化大孔树脂纯化镰形棘豆中总黄酮的工艺 被引量:4

Technology of Purification of Total Flavones from Oxytropis Falcate Bunge with Macroporous Resin by Orthogonal Experiment

摘要 目的采用正交试验法,优选D^(-1)01大孔吸附树脂纯化镰形棘豆中总黄酮的最佳工艺。方法采用紫外分光光度法,以芦丁为对照品,以镰形棘豆中总黄酮回收率为考察指标,对影响总黄酮纯化工艺的因素进行研究;在单因素考察基础上,选择上样液浓度、上样速率、洗脱剂浓度、洗脱速率等4个因素进行正交试验,研究不同因素交互作用及其对总黄酮回收率的影响,确定总黄酮纯化的最佳工艺;测定纯化后的总黄酮纯度,验证纯化效果。结果经正交试验,确定最佳工艺参数:D^(-1)01大孔树脂,上样液浓度为2.75mg·g^(-1),上样速率为1.5ml·min^(-1),洗脱剂浓度为80%,洗脱速率为2.0ml·min^(-1)。在此工艺条件下,镰形棘豆总黄酮的纯度由35.36%提高到69.08%。结论该纯化方法简便可行、重复性好、回收率高,可为大批量纯化镰形棘豆总黄酮提供参考。 Objective To optimize the optimum process of purification of total flavonoids from Oxytropis falcate Bunge by D-101 macroporous adsorptionresin with orthogonal design.Methods With rutin as control,the content of total flavonoids from Oxytropis falcate Bunge was determined with UV spectrophotometry.With the recovery rate of total flavonoids as the main indicator,the factors that affected the purification process of total flavonoids were studied.On the basis of single factor experiments,four factors,including the concentration of samples,loading rate,the concentration of eluent and elution rate,were selected to study the effects of different factors on the recovery rate of total flavonoids.The purity of the purified flavonoids was determined to verify the purification effect.Results The parameters of the optimum process were as follows:D-101 macroporous resin was used,the concentration of the sample solution was 2.75 mg·g-1,the loading rate was 1.5 ml·min-1,the eluent concentration was 80%,and the elution rate was 2.0 ml·min-1.Using this process,the purity of total flavonoids from Oxytropis falcate Bunge was increased from 35.36%to 69.08%.Conclusion This purification method is simple,feasible and reproducible,which can provide reference for industrial purification of total flavonoids from Oxytropis falcate Bunge.
