摘要
采用水提醇沉法提取参芪复方(党参和黄芪药材)中的总多糖。以提取物中的多糖含量和干浸膏得率的总评归一值(OD值)为考察指标,采用Box-Behnken设计结合响应面分析法,考察醇沉浓度、醇沉时间和浓缩比的影响。采用Design Expert 8.0.5b软件对试验结果进行拟合和建模。根据多元二次回归方程的结果,得到优化的醇沉工艺条件为:醇沉浓度75%,醇沉时间13 h,浓缩比1.0(即1 ml浓缩液中含原药材1 g)。照优化条件重复提取3批,结果所得粗多糖提取物中平均多糖含量为25.54%,浸膏得率为31.41%。实测OD值与预测OD值的偏差为-1.72%,说明本试验建立的模型可靠,预测性良好。
Total polysaccharides were extracted from Shenqi compound recipe [a compound of Codonopsis pilosula (Franch.) Nannf. and Astragalus membranaceus (Fisch.) Bge. var. mongholicus (Bge.) Hsiao] by ethanol precipitation process. The effects of ethanol concentration, precipitation time and concentration ratio of ethanol to crud drugs were investigated by Box-Behnken design combined with response surface methodology with overall desirability (OD) value of polysaccharide content and yield of dry extract as an index. The data were analyzed by Design Expert 8.0.5b software. According to the fitted multivariate quadratic regression equation, the optimal process parameters were as follows: ethanol concentration was 75 %, precipitation time was 13 h and concentration ratio was 1 ml : 1 g. The results showed that the mean content of polysaccharide and yield of dry extract in three batches of optimal total polysaccharides extract were 25.54 % and 31.41%. The deviation between measured and predicted values of OD was -1.72 %, indicating the model fitted well.
