摘要
目的:观察气阴双补中药对哮喘大鼠肺组织中转化生长因子-β1(TGF-β1)、Smad2/3表达的影响,研究其对气道重塑的作用机制。方法:以卵清蛋白(OVA)致敏及激发制备慢性哮喘大鼠模型。将48只雄性SD大鼠随机分为正常对照组,哮喘模型组,中药低(5g/kg)、中(10g/kg)、高(20g/kg)剂量组和地塞米松组(0.5mg/kg),每组8只。造模成功后连续给药14d。取肺泡灌洗液(BALF)进行白细胞分类计数,HE染色法观察肺组织病理学改变,计算机图像分析软件测定支气管基底膜周径(Pbm)、气道平滑肌面积(WAm)和气管内壁面积(WAi),逆转录-聚合酶链反应(RT-PCR)检测肺组织TGF-β1 mRNA、Smad2/3 mRNA表达水平。结果:与正常对照组比较,哮喘模型组BALF中白细胞总数及嗜酸性粒细胞百分数明显升高(P<0.05),WAm/Pbm和WAi/Pbm明显增厚(P<0.05),肺组织中TGF-β1、Smad2/3 mRNA表达水平均明显升高(P<0.05);中药低、中、高剂量组和地塞米松组较哮喘模型组上述指标均显著下降(P<0.05)。结论:气阴双补中药能减轻慢性哮喘大鼠肺组织病理学改变,改善气道重塑;其改善慢性哮喘大鼠气道重塑的机制可能与抑制TGF-β1、Smad2/3的表达有关。
Objective: To observe the effects of Chinese herbs with the efficacy of tonifying both qi and yin on the expression of transforming growth factor-β1(TGF-β1) and Smad2/3 in lung tissue of asthmatic rats and explore its underlying mechanism in airway remodeling. Methods: The rat model of chronic asthma was induced by ovalbumin(OVA). A total of 48 male SD rats were randomly divided into 6 groups: the control group, the model group, the low dose group(5g/kg), the medium dose group(10g/kg), the high dose group(20g/kg) and the dexamethasone(DXM) group(0.5mg/kg), with 8 rats in each group. After modeling, the rats in the treatment groups were treated for continuous 14 days. The bronchoalveolar lavage fluid(BALF) was conducted the white blood cell differential count assay. The pathological changes of lung tissues were observed by HE staining. The windpipe basement membrane circumference(Pbm), smooth muscle area of windpipe(WAm) and windpipe internal area(WAi) were measured by the computerized image analysis system. The expression of TGF-β1 and Smad2/3 mRNA in lung tissue were detected by RT-PCR. Results: The OVA-induced chronic asthma significantly enhanced the proportion of leucocytes and eosinophils in BALF(P<0.05), thicken the WAm/Pbm and WAi/Pbm(P<0.05), and obviously increased the expression of TGF-β1 and Smad2/3 mRNA in the lung tissue(P<0.05), all of which were reversed by the treatment of Chinese herbs with the efficacy of tonifying both qi and yin as well as the DXM group(P<0.05). Conclusion: The Chinese herbs with the efficacy of tonifying both qi and yin attenuate the pathological changes of chronic asthmatic rats and improve the airway remodeling, the mechanism of which may relate to its inhibitive effects on the expression of TGF-β1 and Smad2/3.
