摘要
目的:优化铁皮石斛叶多糖的提取工艺。方法:使用闪式提取法对铁皮石斛叶中的多糖成分进行提取,苯酚-硫酸法检测多糖含量,在单因素试验基础上,以料液比、提取时间、提取次数为影响因素,得率为响应值,采用Box-Behnken响应面法,优选最佳提取工艺技术参数。结果:最佳提取工艺条件为以25倍量水提取铁皮石斛叶多糖3次,每次90s,得率为11.52%,与理论得率11.77%接近。结论:经Box-Behnken响应面法优选的铁皮石斛叶多糖的提取工艺,得率与理论值较吻合,模型选用合理有效。
Objective:To optimize polysaccharides extraction technology from Dendrobium officinale Kimura et Migo Leaf.Methods:The homogenate extraction technique of polysaccharides from Dendrobium officinale Kimura et Migo Leaf was used.PhenolSulphuric acid method was employed to detect the content of polysaccharides.On the base of single factor tests,the Box-Behnken Response Surface Analysis was adopted to optimize polysaccharides’ extraction technology technical parameter.Extraction rate was regard as response value and the dosage liquor radio,extraction time,and extraction frequency was done as factors.Results:The optimum extraction conditions were as fallows:extracted polysaccharides 3 times with 25 times the amount of water for 90 seach time.,and the extraction rate was 11.52%,which got close to the theoretical one 11.77%.Conclusion:The adoption of Box-Behnken Response Surface Analysis is reasonable and effective to optimize the extraction technology of polysaccharides from Herba Dendrobii Officinalis Leaf,and the extraction rate coincides with the theoretical one.
