摘要
目的观察人参皂苷Rg1对兔膝关节早期骨性关节炎软骨细胞增殖的影响。方法将50只健康新西兰雄性大白兔随机均分为正常组、模型组和人参皂苷Rg1低、中、高剂量组。采用伸直位管型石膏固定法制备兔膝骨性关节炎早期模型,造模后立即干预,人参皂苷Rg1低(5mg/kg)、中(10mg/kg)、高(20mg/kg)剂量组5 m L/kg体积灌胃给药,正常组和模型组予以等剂量生理盐水灌胃,每周3次,干预6周后取材。采用PCR技术检测P21、Cyclin D1 mRNA表达,Western-blot技术检测相应蛋白的表达,PCNA法检测软骨细胞增殖活性。结果人参皂苷Rg1低、中、高剂量组P21 mRNA及相应蛋白的表达均低于模型组(P<0.05或P<0.01),而Cyclin D1 mRNA及相应蛋白的表达高于模型组(P<0.05或P<0.01),PCNA检测显示人参皂苷Rg1中、高剂量组软骨细胞增殖率明显高于模型组(P<0.05)。各项指标中以中剂量组效果最佳,即存在一定的量效关系。结论人参皂苷Rg1可有效提高软骨细胞G1期调节蛋白Cyclin Dl的表达,抑制P21表达,从而促进软骨细胞增殖,对软骨组织有着积极的修复作用。
Objective To observe the influences of Ginsenoside Rg1 on chodrocyte proliferation of early knee osteoarthritis( OA) rabbits. Methods 50 healthy adult New Zealand rabbits were equally divided into normal group,model group and Ginsenoside Rg1 low,medium and high dose groups randomly. 40 rabbits were established models with plaster cast unilateral hindlimb straighten place braking method,intervention was used at the beginning of model preparation. Ginsenoside Rg1 low( 5mg/kg),medium( 10 mg/kg) and high( 20 mg/kg) dose group were gastriced by 5 m L/kg,meanwile,the normal group and model group were gastriced with isodose physiological saline,3 times a week,interent materials after 6 weeks. Real- Time PCR method was used to detect the expression of P21,Cyclin D1 mRNA,Western- blot method was used to detect the corresponding proten,cartilage cell proliferation activity was detected by PCNA. Results The expression of P21,mRNA and corresponding proten of Ginsenoside Rg1 low,medium and high dose group were lower than model group( P〈0. 05 or P〈0.01),Cyclin D1,mRNA and corresponding proten were higher than model group( P〈0. 05 or P〈0. 01),PCNA showed that the proliferation rate of chondrocytes of Rg1 medium and high dose groups were higher than model group( P〈0. 01). There existed a certain dose- effect relationship,of which the effect of medium dose group was the best. Conclusion Ginsenoside Rg1 can effectively increase the expression of the G1 phase protein of Cyclin D1 and inhibit the expression of P21,thus to promote cartilage cell proliferation,it plays a positive role in cartilage repairing.
