摘要
目的观察人参皂苷Rg1、Rb1对体外培养软骨细胞,IL-1诱导软骨细胞凋亡的影响,并比较其对细胞影响的差异性。方法成功培养出软骨细胞后,进行人参皂苷影响细胞代谢浓度筛选,设人参皂苷Rg1不同浓度组、人参皂苷Rb1不同浓度组,MTT法检测人参皂苷对软骨细胞代谢的影响,选取其Rg1、Rb1合适浓度作为干预细胞凋亡浓度组,设立人参皂苷Rg1、Rb1组和对照组,IL-1诱导软骨细胞凋亡,流式细胞仪检测其凋亡数量和比例,透射电镜检测细胞凋亡形态和亚微结构。结果人参皂苷Rg1m、Rb1m组对软骨细胞调亡有明显的抑制作用,在抑制软骨细胞过度凋亡无明显差异。结论人参皂苷Rg1、Rb1能明显抑制软骨细胞过度的调亡,抑制膝骨性关节炎的发生、发展,为进一步在体实验提供理论基础。
Objective To observe the impacts of ginsenoside Rgl and Rbl on chondrocyte cultivated in vitro and chondrocyte apoptosis induced by IL - 1 ,arid compare the differences of them in the impacts on cells. Methods After the successful culture of chondrocytes,ginsenosides were used to impact the con- centrations of cell metabolism. The screening was performed. Ginsenoside Rgl and Rblgroups of different concentrations were established respectively. MTT was adopted to determine the impacts of ginsenosides on chondrocyte metabolism. Rgl and Rbl of suitable concentrations were selected to be the intervention apoptosis concentration groups. Ginsenoside Rgl and Rbl groups and control group were set up. Chondrocyte apoptosis was induced by IL - 1. Flow cytometry was applied to determine the numbers of apoptosis and proportion. Transmission electron microscope was used to determine the morphology of apoptosis and ultramicrostructure. Results Ginsenoside Rglm and Rblm groups presented apparent inhibition on the apoptosis of chondrocyte. But there was no apparent difference in the inhibition on excessive apoptosis of chondrocyte in comparison. Conclusion Ginsenoside Rg1 and Rbl inhibit apparently excessive apoptosis of chondrocyte and suppress the occurrence and development of osteoarthritis of knee ,which provides the theoretic basis for a further experiment in vivo.
