摘要
运用BIOLOG技术研究了猫儿山亚热带常绿阔叶林不同土层深度的土壤微生物群落功能多样性。结果表明不同土层深度的土壤微生物群落功能多样性差异显著。土壤颜色变化率(AWCD)随培养时间延长而逐渐增加,但AWCD值随着土层的加深而逐渐降低。不同土层土壤微生物对6类碳源的利用能力基本一致,排序为:氨基酸类>酯类>胺类>酸类>醇类>碳水化合物类,随着土壤深度的加深,微生物利用各类碳源的能力呈下降趋势,氨基酸类、酯类和胺类为不同土层土壤微生物的主要碳源。土壤微生物群落Shannon-Weinner指数、丰富度指数和均匀度指数也随着土壤深度的加深呈下降趋势。主成分分析结果表明,从31个因素中提取的与碳源利用相关的主成分1、主成分2分别能解释总变异的33.32%和23.14%,在主成分分析中起主要贡献作用的是碳水化合物类、酯类和胺类碳源。土壤理化性质与土壤微生物群落功能多样性的相关性分析结果表明,总有机碳、速效氮、速效磷和速效钾是造成不同土层土壤微生物群落多样性差异的主要原因。
BIOLOG analysis was used to study the spatial heterogeneity in different soil layers of soil microbial functional diversity in subtropical evergreen broad-leaved forest of Maoer Mountain. The results showed that there were significant differences in the functional diversity of microbial communities between soil layers. The average well color development (AWCD), which directly reflected microbial activity and functional diversity, increased over time, while AWCD decreased along with the increasing depth of soil layers. The utilization of carbon sources of different soil layers was in the same order of amino acids〉esters〉amines〉 acids〉alcohols〉carbohydrates. The ability of soil microbial organisms to metabolize carbon sources decreased with increasing soil depth, with amino acids, esters and amines being the main carbon sources. The Shannon-Weinner, richness and evenness indexes of soil microbial communities also decreased along with increasing depth of soil layers. The principal component analysis (PCA) identified two main factors which explained 33.32% and 23.14% of the total variation of the carbon utilization profiles. The carbon sources utilized by soil microbial communities were different among soil layers. Carbohydrates, esters and amines mainly contributed to the variation in the two principal components. The correlation analysis of soil physical-chemical properties and microbial functional diversity showed that total organic carbon, available nitrogen, available phosphorus and available potassium were the primary drivers of soil microbial communities in different soil layers.
