红外光谱在滇龙胆干燥方法选择上的应用 被引量:1

摘要 目的筛选滇龙胆的最佳干燥方法。方法采用傅里叶变换红外光谱法(Fourier transform infrared spectroscopy,FTIR)结合多元统计分析对晒干(GZCK)、发汗干燥(FZ)、45℃烘箱干燥(HZ45)、微波干燥(WZ)及室内阴干(YG)5种干燥方法处理的45份滇龙胆样品进行分析。结果精密度、稳定性及重现性试验的匹配值在99.33~99.90之间,RSD≤0.01%,表明该法稳定可靠。谱峰比较发现,晒干处理样品在龙胆苦苷的两个主要特征吸收峰1 611和1 075 cm-1附近吸光度值比其它干燥处理的高,表明晒干有利于滇龙胆中龙胆苦苷的保留。原始光谱经自动平滑+自动基线校正+Norris平滑(5∶5)+二阶求导预处理并进行偏最小二乘判别分析(Partial least square discriminant analysis,PLS-DA),结果显示同一干燥处理的样品相对聚集,判别效果较好。谱图检索显示相同干燥方法处理的样品匹配正确率为93.3%,表明该法用于分析不同干燥方法的滇龙胆样品效果较理想。结论晒干是滇龙胆的最佳干燥方法。该法可直观有效地评价中药干燥方法的优劣。 Objective To screen the best drying methods of Gentiana rigescens. Methods Fourier transform infrared (FTIR) spectroscopy combined with multivariate analysis was used for analyzing 45 samples of G. rigescens which were treated by five dr- ying methods including sun- drying, sweating, drying in an oven at 45 ℃ , microwave drying and natural drying in the shade. Re- suits The matching values of precision, stability and repeatability were from 99.33% to 99.90% , and RSD was less than 0.01% , which showed that the method was stable and reliable. By comparing the spectral peaks,it can be found that the absorbance of sam- ples processed by sun - drying was higher than that of others at the two main characteristic absorption peaks of gentiopieroside with the wave number of 1 611 and 1 075 cm^-1. It indicated that sun - drying made a contribution to maintaining the gentiopicroside of G. rigescens samples. The original infrared spectra data were preprocessed by automatic smoothing, automatic calibration, Norris smoothing (5: 5 ) and the second derivative. Then the processed data were used for partial least square discriminant analysis ( PLS - DA). The results of PLS - DA showed that samples with the same drying methods could be grouped and the classification of this method was the best. Spectral retrieval results showed the accuracy rate of the same drying samples was 93.3%. It could be used for analyzing G. rigescens with different drying methods and could also reach the desired results. Conclusion Sun - drying was recognized as the best method of G. rigescens samples. This method can intuitively and effectively evaluate the advantages and dis- advantages of the drying methods of Chinese medicines.
