摘要
目的研究健脾化瘀方对体外模拟缺氧微环境下结肠癌SW480细胞生物学行为的影响。方法 CoCl2化学模拟结肠癌SW480细胞缺氧微环境,实验分为空白组、CoCl2组、健脾化瘀方不同浓度(0.2、0.4、0.8mg/mL)处理组。采用MTT法检测不同浓度健脾化瘀方对缺氧条件下结肠癌SW480细胞株增殖能力、黏附能力的影响,Transwell小室法观察对SW480细胞侵袭能力的影响。结果缺氧条件下SW480细胞增殖能力有所下降,48h时,中、高浓度健脾化瘀方对缺氧条件下SW480细胞的增殖有明显抑制作用,与CoCl2组比较,差异有统计学意义(P<0.05,P<0.01);不同浓度的健脾化瘀方作用于缺氧条件下SW480细胞后,细胞黏附能力明显下降,并呈浓度依赖性;CoCl2组较空白组侵袭细胞数明显增多,各浓度的健脾化瘀方均能抑制SW480细胞体外侵袭能力,各浓度组穿过小室的细胞数随健脾化瘀方浓度的增大而减少,侵袭抑制率增大,差异有统计学意义(P<0.01)。结论健脾化瘀方在体外可明显抑制缺氧微环境下SW480细胞增殖、黏附及侵袭能力,具有一定的浓度依赖性。
Objective To study the influence of Jianpi Huayu Formula on the biological behavior of colon cancer SW480 cells under in-vitro simulative hypoxic microenvironment.Methods Chemical solution CoCl2 was used to simulate the hypoxic microenvironment of colon cancer SW480 cells.This study was divided into CoCl2 group,blank group and 3treatment groups with different-concentrations of Jianpi Huayu Formula（0.2,0.4and 0.8mg/mL）.Methyl thiazolyl tetrazolium（MTT）was applied to detect the influence of Jianpi Huayu Formula on the proliferation and adherence of colon cancer SW480 cell strains under hypoxic microenvironment.Transswell was utilized to observe the influence of Jianpi Huayu Formula on the invasion of SW480 cells.Results Under hypoxic microenvironment,the proliferation of SW480 cells decreased evidently,and 48 hafter treatment,the high-and middle-concentrations of Jianpi Huayu Formula had obvious inhibiting effect on the proliferation of SW480 cells,and the difference was significant when compared with CoCl2group（P〈0.05,P〈0.01）.The adherence of SW480 cells decreased after the administration of Jianpi Huayu Formula in a concentration-dependent manner.The number of invasive cells was markedly higher in CoCl2 group than in blank group,and Jianpi Huayu Formula had suppressive effects on the in-vitro invasive ability of SW480 cells.The number of cells passing the Transwell small chamber decreased along with the increased concentration of Jianpi Huayu Formula,and the inhibition rate on invasion was greatly increased（P〈0.01）.Conclusion Jianpi Huayu Formula can significantly inhibit the proliferation,invasion and metastasis of SW480 cells in a concentration-dependent manner.
