摘要
目的:研究C/EBPα蛋白对结肠癌组织中癌细胞的作用。方法:收集2015年9月-2017年9月我院确诊为结肠癌60例患者的临床资料,检测患者癌组织及癌旁正常组织中的C/EBPα表达,对比C/EBPα低表达及高表达之间存在的病理参数差异,观察低表达C/EBPα与高表达C/EBPα对癌组织侵袭相关蛋白产生的影响。结果:癌旁正常组织低表达C/EBPα率低于癌组织,差异有统计学意义(P<0.05);低表达C/EBPα与高表达在性别及年龄方面对比,差异无统计学意义(P>0.05);低表达C/EBPα肿瘤直径长,且肿瘤TNM分期晚,比较差异有统计学意义(P<0.05);经Western blot实验发现,C/EBPα低表达患者的肿瘤直径大,肿瘤TNM分期晚,而高表达C/EBPα患者SW480细胞MMP-2、MMP-9、KLF5表达越低,E钙蛋白表达越高,划痕实验指出,在48h后,低表达C/EBPα的SW480细胞迁移率为53.1%,高于高表达的22.51%。结论:低表达C/EBPα同结肠癌患者的肿瘤TNM分期及肿瘤转移之间有着密切相关性,C/EBPα高表达利于降低结肠SW480细胞的侵袭性,可作为未来结肠癌临床治疗研究的重要方向。
Objective:To study the effect of C/EBPαprotein on cancer cell in colon cancer tissue.Methods:The clinical data of 60 patients confirmed as colon cancer in our hospital from September 2015 to September 2017 were collected.The C/EBPαexpression in cancer tissue and adjacent normal tissue of patients were detected.The pathological parameters difference between the low-expression and high-expression C/EBPαwere compared.The effects of low-expression and high-expression C/EBPαon cancer cell invasive protein were observed.Results:The low-expression C/EBPαrate of cancer adjacent normal tissue was lower than that of cancer tissue(P<0.05);there was no statistical difference in the gender and age between the low-expression and high-expression C/EBPα(P>0.05);the tumor diameter of low-expression C/EBPαwas long,and tumor TNM staging was late,with statistical difference(P<0.05);Western blot test showed that:the tumor diameter of low-expression C/EBPαpatients was larger,the tumor TNM staging was later,while the MMP-2,MMP-9,KLF5 expression of SW480 cell in high-expression C/EBPαpatients were lower,the E-cadherin expression was higher;the scratch test showed that:after 48h,the migration rate of SW480 cell in low-expression C/EBPαwas 53.1%,which was higher than 22.51%in high-expression C/EBPα.Conclusion:The low-expression C/EBPαhas close relation with tumor metastasis and TNM staging of colon cancer patients,and the high-expression C/EBPαcan decrease invasion of SW480 cell,which can be taken as the important direction of future research on the clinical treatment of colon cancer.
