HPLC-MS技术研究配伍对四逆汤中附子生物碱特征图谱的影响 被引量:3

Research on Compatibility of the Specific Chromatogram of Alkaloids from Aconite in “Sini Decoction” by HPLC-MS

摘要 目的:采用HPLC-MS技术比较配伍甘草前后四逆汤中附子生物碱的特征峰在量和质上的异同,阐明配伍对四逆汤中附子生物碱特征图谱的影响。方法:应用HPLC法建立四逆汤中附子生物碱的特征图谱,考察该方法的可靠性,测定附子、缺附子四逆汤、四逆汤中生物碱的特征图谱,并采用HPLC-MS检测推测特征峰的组成。结果:所建立的HPLC测定方法稳定、可靠、精密度好,可用于四逆汤中附子生物碱特征图谱的研究;以苯甲酰新乌头原碱为参比峰,标定了18个特征峰,其中8个特征峰来源于附子,8个特征峰来源于干姜和甘草水煎液,2个特征峰共同来源于二者;配伍后来源于附子的10个特征峰峰面积均有不同程度降低,甚至特征峰消失;结合文献推测来源于附子的10个特征峰。结论:配伍对四逆汤中来源于附子的特征峰有较为显著的影响,其溶出量显著降低。 Objectlve:To study the effect of combination of glycyrrhiza on the specific chromatogram of alkaloids from Aconite in "Sini Decoction" by HPLC- MS. Methods : A method of HPLC specific chromatogram of alkaloids from Aconite in "Sini Decoction" was established. The precision and stability of the method was measured in Radix Aconiti Lateralis Preparata, "Sini Decoction" without Radix Aconiti Lateralis Preparata and "Sini Decoction". Then the structures of specific peaks were speculated with HPLC-MS. Results: The HPLC method is stable, reliable and with a good reproducibility which could be used to determine the specific chromatogram of alkaloids from Aconite in "Sini Decoction'. Eighteen peaks were selected as specific peaks with benzoylmesaconine peak as the reference peak. Eight of them were from Aconite, 8 of them were from "" Sini Decoction" without aconite and the other 2 peaks were from both of them. The areas of specific peaks of alkaloids from Aconite in " Sini Decoction" were smaller even disappeared after the compatibility. Combined with. literature data, ten chromatographic peaks were inferred. Conclusion : The specific chromatogram of alkaloids from Aconite in " Sini Decoction" is markedly influenced by its compatibility and the dissolving-out quantity is reduced obviously.
