摘要
目的:探讨自体骨髓基质干细胞(BMSCs)移植联合表皮生长因子受体(EGFR)抑制剂对大鼠脊髓损伤(SCI)治疗的效果。方法:培养与纯化骨髓基质干细胞。60只SD大鼠制备脊髓损伤模型,随机分为4组:基质干细胞组、EGFR抑制剂组、联合组和对照组。采用BBB法进行运动功能评分和切片观察来检测修复情况。结果:随着时间的延长,四个组的大鼠的运动功能都是逐渐恢复,术后第7天起,对照组BBB评分明显低于其余三个组(P<0.05),且联合组大鼠的神经功能恢复好于基质干细胞组和表皮生长因子受体抑制剂组(P<0.05)。组织学染色显示三个实验组脊髓组织恢复均好于对照组(P<0.05);联合治疗组脊髓组织恢复好于基质干细胞组与表皮生长因子受体抑制剂组(P<0.05)。结论:骨髓基质干细胞联合EGFR抑制剂具有促进大鼠脊髓损伤后神经功能修复的作用。
Objective:To evaluate the effects of transplantation of bone marrow stromal cells (BMSCs) with inhibitors of the epider-mal growth factor receptor ( EGFR) therapy after rats spinal cordinjury ( SCI).Methods:BMSCs were cultured and purified in vitro . The rat model was prepared with modified Alle method .Sixty rats were devided into four groups:stem cell group , inhibitors of EGFR group, combination group and control group.The motor function was evaluated with BBB score and the neurological function recovery were observed .Results:Statistical differences were showed in inter group.Compared with control group , in practical group ,the motor function and morphological recovery was obvious in MSCs combined EGFR inhibitors group .Conclusion:The neurological function of SCI rats can be obviously improved by BMSCs transplantation combined with Inhibitors of the epidermal growth factor receptor .
