枸杞多糖诱导大鼠骨髓间充质细胞向神经元样细胞转化的实验研究 被引量:10

Experimental study on transformation of rat bone marrow mesenchymal stem cells into neuron-like cells induced by Lycium barbarum polysaccharide

摘要 ①目的探讨枸杞多糖诱导骨髓间充质细胞(BMSCs)向神经细胞转化的可能机制。②方法用贴壁筛选法分离培养出贴壁生长良好的骨髓BMSCs。选取第3代细胞,当传代细胞长满瓶底的80%以上时,先用含15%FBS和1g/L枸杞多糖的DMEM培养基预诱导24小时,然后,用不含血清的1g/L枸杞多糖的DMEM培养基诱导。光镜下观察细胞形态;用免疫组化技术检测细胞Nes-tin、NF、GFAP的表达。③结果预诱导后,BMSCs没有变化。而经过枸杞多糖诱导后,细胞在4小时后即出现形态学上的改变,细胞变凸,立体感增强,从胞体伸出突起。免疫组化检测显示,细胞Nestin、NF染色阳性,GFAP阴性。④结论骨髓间充质细胞经过枸杞多糖的诱导可以转化为神经细胞。 Objective To study the possibility that lycium barbarum polysaccharide (LBP) induced differentiation of adult rat bone marrow mesenchymal stem cells (BMSCs) into neurons. Methods BMSCs from adult rat were isolated and cultured. Then BMSCs were induced by lycium barbarum polysaccharide in serum - free medium for different period. BMSCs were pretreated with preinduction medium consisting of DMEM 15%, FCS 1g/L, LBP for 24 hours, then induced with induction medium consisting of DMEM 1g/L and LBP. The morphological changes of the cells were observed under phase contrast microscope. The neuronal specific markers, neurofilement(NF) , nestin , and gliall fibriUary acaidic protein(GFAP) were evaluated by indirect immunocyto - chemistry staining. Results After induction for 4 hours , some showed morphological changes obverously. Twenty - four hours later, the differentiated cells showed typoal neuronal morphology. The expression of NF and nestin was positive but GFAP was negative. Conclusion Lycium barbarum polysaccharide has inductive effect on differentiation of BMSCs into neurons of adult rat in vitro.
