摘要
目的研究茵陈蒿汤对ABO血型抗原免疫小鼠肿瘤坏死因子-α(TNF-α)分泌水平的影响及TNF-α与抗-A水平变化之间的相关性。方法以人A型红细胞免疫BALB/c小鼠为模型,通过ELISA检测小鼠外周血TNF-α水平变化,应用间接抗球蛋白试验,吸收放散试验检测小鼠血清IgG型抗-A,观察A型红细胞免疫组和茵陈蒿汤治疗组中BALB/c小鼠体内TNF-α水平变化及与IgG型抗-A水平之间的相关性。结果对照组、A型红细胞免疫组和茵陈蒿汤治疗组TNF-α检测结果分别为:(83.50±6.32)ng/L、(135.56±44.14)ng/L和(109.44±12.50)ng/L,组间比较存在统计学差异(P<0.05),组间两两比较P值均<0.05;茵陈蒿汤治疗组有效率为61.11%,有效组和无效组之间TNF-α含量比较无统计学差异;TNF-α水平与抗-A水平之间呈显著性正相关关系(P<0.05)。结论小鼠动物实验提示茵陈蒿汤可抑制TNF-α和ABO血型抗体的分泌,TNF-α水平和ABO血型抗体水平之间的变化呈正相关。
Objective To research effects of Yinchenhao decoction on secretion of tumor necrosis factor-α( TNF-α) in mice immunized with ABO blood group antigens and to investigate the correlation of TNF-α level with the changes of anti-A. Methods Taking BALB/c mice immunized with human erythrocytes of A blood group as a model,the TNF-α in BALB/c mice plasma was detected with ELISA,and the IgG-typed anti-A of BALB/c mice was assayed with indirect antiglobulin test( IAT) as well as absorption and elution test. The correlation of TNF-α level with the changes of IgG-typed anti-A,in immune group treated with A-type erythrocytes and therapy group treated with Yinchenhao decoction,was investigated. Results TNF-α in control group,A-type erythrocyte immune group and Yinchenhao decoction treatment group were( 83. 50 ± 6. 32) ng/L,( 135. 56 ± 44. 14) ng/L and( 109. 44 ± 12. 50) ng/L,respectively. There existed significant differences between groups( P 〈0. 05),and showed all P〈 0. 05 by multiple comparisons. The effective rate in Yinchenhao decoction treatment group was 61. 11%,and there showed no significant differences of TNF-α levels in effective group than in ineffective group. There showed a significant positive correlation between the TNF-α levels and the changes of anti-A( P〈 0. 05). Conclusion BALB/c mouse study suggested that Yinchenhao decoction can inhibit the secretion of TNF-α and ABO blood group antibodies,and there showed a significant positive correlation between the levels of TNF-α and the changes of anti-A.
