摘要
利用大鼠离体肝脏灌流技术以及大鼠原代培养的肝细胞研究了大豆黄酮 (Da)对SD大鼠肝脏氮代谢和IGF 1生成的影响。结果表明 ,Da可使大鼠离体肝脏灌流液中尿素氮浓度下降 ,并有一定的剂量效应和时间效应。在给药后灌流的1,2 ,3和 4h内 ,与对照组相比 ,1× 10 -4mol·L-1的Da使大鼠离体灌流的肝脏产生的尿素氮分别下降了 13 6 4 % ,2 3 18% ,34 0 2 % (P <0 0 1)和 32 91% (P <0 0 1)。 4× 10 -5mol·L-1的Da对大鼠肝脏尿素氮生成影响具有类似但较微弱的效应。Da还可在一定程度上抑制大鼠离体灌流的肝组织中谷丙转氨酶 (GPT)的活性。 1× 10 -5mol·L-1的Da可使3 H 亮氨酸的参入量与对照组相比提高了 2 1 38% (P <0 0 5 )。Da对离体灌流的大鼠肝脏胰岛素样生长因子 1(IGF 1)的水平也有微弱的促进作用。提示Da可通过增加大鼠肝脏氮的储留 ,促进大鼠肝细胞蛋白质的合成以及在一定程度上影响肝脏GPT的活性和IGF 1水平 ,而调节肝脏的氮代谢。
The technique of isolated perfused rat liver in vitro and cultured rat hepatocytes were used to study the effects of daidzein on liver nitrogen metabolism and insulin like growth factor 1(IGF?1)level of Sprague Dawley rat.The results showed that urea?nitrogen concentration of perfused medium was significantly affected by daidzein in a dose?dependent and time?dependent manner.The urea?nitrogen level was decreased by 13 64%,23 18%,34 02%( P <0 01)and 32 91%( P <0 01)for 1,2,3 and 4 h after administering daidzein at the dose of 1×10 -4 mol·L -1 ,respectively,compared to that of control.The daidzein at the dose of 4×10 -5 mol·L -1 had the similar effect on urea?nitrogen level.Glutamic?pyruvic transaminase(GTP)activity of isolated perfused rat liver was inhibited by daidzein,but the difference was not significant compared with control.The protein synthesis of rat hepatocyte was enhanced by daidzein,the content of 3 H?leucine incorporation into rat hepatocyte was significantly increased by 21 38%( P <0 05)with daidzein?treated(1×10 -5 mol·L -1 ).IGF?1 level of rat hepatocyte were also affected by daidzein,but no significant difference was found compared to control.It is suggested that daidzein can regulate nitrogen metabolism of rat liver by means of increasing nitrogen retention,enhancing synthesis of protein and affecting GTP activity and IGF?1 level of rat liver.
