大分子物质对中药注射剂外观颜色的影响 被引量:9

摘要 目的:探究大分子物质对中药注射剂外观颜色的影响。方法:取清开灵、双黄连、丹参和灯盏细辛4种中药注射液,用3、10、30 k超滤管截留制备去大分子注射液和富含大分子注射液,用Tecan酶标仪扫描紫外-可见吸收曲线,分析曲线下面积的变化;用HPLC法检测并分析10 k去大分子液的指纹图谱变化,用冷冻干燥法检测固体物含量。然后用10 k分子筛过滤4种注射液,反复洗涤截留液获得大分子物质。最后用焦性没食子酸和鞣酸模拟中药注射剂变色过程。结果:4种注射液用不同超滤管超滤后,去大分子液的颜色明显变浅,吸收曲线下面积变小,且截留分子量越小,去大分子液的吸收曲线下面积也越小;富含大分子部分的颜色则明显变深,曲线下面积增大;10 k去大分子液的指纹图谱与原液相似,指纹图谱总峰面积变化较小,固体物含量波动在5%以内;焦性没食子酸和鞣酸溶液变色后去除大分子物质,颜色也明显变浅。结论:大分子物质是中药注射剂有色物质的重要来源,去除大分子物质可改善中药注射液外观质量。 Objective: To verify the influence of macromolecule on appearance quality of Traditional Chinese Medicine Injections(TCMIs). Methods: Macromolecule-free and macromolecule-rich TCMIs were obtained by filtering four TCMIs, Qingkailing Injection(清开灵注射液), Shuanghuanglian Injection(双黄连注射液), Danshen Injection(丹参注射液), and Dengzhanxixin Injection(灯盏细辛注射液), through molecular sieves with aperture of3 k, 10 k or 30 k. Their ultraviolet-visible absorption spectra were scanned with a Tecan multi-plate reader, and areas under the absorption spectra were calculated. HPLC fingerprints of macromolecule-free TCMIs made by 10 k molecule sieves were determined, all the peak areas were calculated, and the solids in the macromoleculefree TCMIs were weighted. As for macromolecule-rich TCMIs made by 10 k molecule sieves, the macromolecules in them were rinsed, freeze-dried and weighted. Pyrogallic acid and tannic acid were introduced to mimic TCIMs’ photochromic reactions. Results: The color of TCMIs was lightened and the areas under absorption spectra decreased in macromolecule-free TCMIs. As for macromolecule-rich TCMIs, their color was darkened and the areas under absorption spectra increased. The fingerprint of 10 k macromolecule-free TCMI was similar to that of the original TCMI, and the sum of peak-areas and the solids of 10 k macromolecule-free TCMI were also similar to those of original TCMI. Pyrogallic acid and tannic acid easily became dark in weak alkaline solution, solids content fluctuates below 5%. However, the solution color was lightened when macromolecules were removed with molecular sieves. Conclusion: Macromolecules are responsible for the color of TCMIs, and macromolecule removal improves the appearance quality of TCMIs.
