摘要
研究中药抗癌与抑制血管形成之间的关系。应用细胞形态学、细胞生长曲线、MTT检测、DNA琼脂糖凝胶电泳、流式细胞仪检测方法等。结果表明去甲斑蝥素对体外培养的人脐静脉内皮细胞有显著的生长抑制作用 ,药效呈时间依赖性 ,并在一定范围内 (5~ 80 μmol/L)与剂量呈正相关 ,半有效量为 4 0 μmol/L。形态学观察证明给药后 2 4h ,坏死细胞数随给药浓度增多。分离提取细胞DNA ,在琼脂糖凝胶电泳后可见弥散状核酸降解带 ,未见典型的细胞凋亡梯状带。流式细胞仪检测表明 ,随着给药浓度增高 ,四倍体细胞数显著增多 ,不见明显的凋亡细胞峰。表明去甲斑蝥素对人脐静脉内皮细胞有较强的细胞毒作用 ,并能抑制细胞正常分裂。
The inhibitory effects of norcantharidin on neoplastic angiogenesis were investigated by cell morphological observation, drawing of cell growth curve, MTT assay, DNA agarose gel electrophoresis and flow cytometry. The results showed that norcantharidin exhibited a marked inhibitory effect on the growth of human umblilical venous endothelial cells in vitro, in both a time effect manner and a dose effect manner with IC50 equal to 40 μmol/L, and to a certain extent (5~80 μmol/L), there was a positive correlation between the effect and doses. The results of cell morphological observation showed that 24 hours after giving norcantharidin, the number of necrotic cells increased in a concentration effect manner, and the DNA of the cells exhibited diffuse nucleic acid degradation bands but no typical apoptosis bands after agarose gel electrophoresis. Flow cytometry showed that the number of tetraploid cells markedly increased after giving norcantharidin in a concentration effect manner, but there were no remarkable apoptosis peaks. All the results imply that norcantharidin has remarkable cytotoxic effects and can inhibit normal cell division and the growth of human umblilical venous endothelial line in vitro.
