摘要
目的:观察疏肝健脾方不同提取物对CCl4诱发的化学肝损伤小鼠的保护作用。方法:将小鼠随机分为正常组、模型组、疏肝健脾方水提组、醇提组、醇水双提组、联苯双酯(BP)组,灌胃给予不同工艺制备的提取物,每天一次,连续10d,末次给药8h后,按0.1ml/10g一次性注射0.1%CCl4溶液复制小鼠化学性肝损伤模型。分光光度法测定血清中谷氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)及肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)含量;HE染色观察肝病理组织学改变;免疫组化染色检测NF-κB、TNF-α蛋白的表达。结果:疏肝健脾方水提取物(15.6g/kg)、醇提取物(15.6g/kg)、醇水双提取物(15.6g/kg)不仅能降低ALT、AST、MDA含量,升高SOD、GSH-Px活性,还能减轻肝脏病理损伤程度,抑制肝脏NF-κB、TNF-α蛋白的表达。结论:疏肝健脾方对CCl4诱发的肝损伤小鼠有一定的保护作用,其机制可能与抗机体脂质过氧化有关。
Objectives: To study the protective effect of Shuganjianpi Formula different extracts on carbon tetrachloride-induced chemical liver injury. Methods: The mice are randomly divided into six groups: the control,the model,the Shuganjianpi Formula Water extraction group,the alcohol extraction group,the alcohol water double extraction group( 15. 6g /kg),The Biphenyldicarboxylate( 0. 1g /kg) group. The treatment group were given by lavage,one time a day for 10 days. After being given drug for 8 hours,the model were induced by injection of carbon 0.1% tetrachloride-olive oil solution( 0. 1ml /10g) by intraperitoneal injection. After being given drug for 10 days,ALT,AST,SOD,MDA,and GSH-Px were examined by colorimetric methods. The histopathologic changes of the hepatic tissues were observed by the HE stained method. Immunohistochemical staining were used to detect myocardial related protein including TNF-α and NF-κB,respectively. Results:Shuganjianpi Formula different extracts could decrease the ALT,AST and MDA level. Shuganjianpi Formula different extracts could increase the SOD and the GSH-Px level. the degree of the liver injury were also inhibited by Shuganjianpi Formula different extracts. The expressions of TNF-α and NF-κB p65 of model group were significantly inhibited. Conclusions: Shuganjianpi Formula different extracts have certain protective effect on chemical hepatic injury in mice,and its mechanism might be related with resisting lipid peroxidation. Among them,Alcohol water double extraction group could significant reduce the degree of liver injury.
