摘要
以提取的无芒雀麦基因组DNA为模板,通过试验筛选出了ISSR-PCR反应体系中最适宜的各组分浓度,并用POPGENE软件对数据进行分析。结果表明,供试25份无芒雀麦材料总的Nei's基因多样性指数为0.2434,Shannon信息指数为0.3721;聚类分析研究将供试25份材料分为五大类,第一类为编号1、4、11、14、15、18、19、21、23和15号材料,第二类为编号2、5、8、9、10、22和24号材料,第三类为编号3、6和17号材料,第四类为20号材料,第五类为7、12和16号材料。
Template DNA for ISSR was extracted fromBromus inermis leaf tissue,ISSR amplification system was optimized,an optimal reaction system was established,data were analyzed with the software POPGENE.The results showed that the total Nei's gene diversity index of 5 Bromus inermis Leyss accessions was 0.2434 and Shannons Information Index was 0.3721;25 materials were divided into four groups, group one was consist of ten Bromus inermis materials:No.1,No.4,No.11,No.14,No.15,No.18,No.19, No.21,No.23 and No.25,group two was composed of eight materials:No.2,No.5,No.8,No.9,No.10, No.22 and No.24,group three includes three Bromus inermis materials:No.3,No.6 and No.7,group four included only one Bromus inermis materials:No.20,and the last group had three materials:No.7,No.12, and No.16.
