miR-151-3p在原发性肾病综合征患儿血清中的表达及临床意义 被引量:1

摘要 目的检测原发性肾病综合征(primary nephrotic syndrome,PNS)患儿血清中miR-151-3p的表达水平,探讨其在PNS治疗中的临床意义。方法收集我院2011-2012年住院的PNS患儿40例,根据对糖皮质激素治疗的反应分为激素敏感型(steroid-sensitive nephrotic syndrome,SSNS)组(30例)和激素抵抗型(steroid-resistant nephrotic syndrome,SRNS)组(10例)。同时,取年龄、性别匹配的健康儿童20例作为健康对照组。利用实时定量PCR检测3组在治疗前后血清和尿液中miR-151-3p的表达水平。分析血清miR-151-3p与PNS临床指标的相关性。结果与健康对照组相比,PNS患儿血清中miR-151-3p的表达水平升高(P<0.05)。SSNS组治疗后血清中miR-151-3p的表达水平较治疗前下降(P<0.05)。PNS患儿尿液中miR-151-3p的表达水平无明显变化(P>0.05)。血清中的miR-151-3p水平与24 h尿蛋白量、血清胆固醇含量呈正相关(P<0.05),而与患儿的年龄、性别以及血浆中白蛋白含量无相关性(P<0.05)。结论 PNS患儿血清中miR-151-3p的高表达提示其具有作为PNS患儿治疗靶标的潜能。 Objective To examine miR-151-3p syndrome (PNS) and explore its clinical value. Methods serum expression in children with primary nephrotic Forty children with PNS who were admitted into our hospital from 2011 to 2012 were investigated. According to the response to the effect of glucocorticosteroid, 30 children were assigned into a steroid-sensitive nephrotic syndrome (SSNS) group and 10 into a steroid-resistant nephrotic syndrome (SRNS) group. Twenty age and sex matched healthy children were enrolled as healthy controls. Real-time PCR was performed to detect the expression of serum miR-151-3p. Then we analyzed the miR-151-3p expression in SSNS group before and after steroid treatment. We also investigated miR-151-3p expression in urine samples, Finally, we analyzed the relationship between miR-151-3p expression and the clin- ical characteristics of PNS. Results Compared to the healthy children group, miR-151-3p was upregulated in the PNS groups (P 〈 0. 05). MiR-151-3p expression was reduced after steroid treatment in the SSNS group (P〈0. 05). There was no significant difference in miR-151-3p expression from urine samples of PNS and healthy children (P 〉 0. 05 ). There existed a positive correlation between serum miR-151-3p expression and se-rum cholesterol and urinary protein of 24 hours, but no correlation between serum miR-151-3p expression and serum albumin. Conclusion MiR-151-3p expression is upregulated in blood serum of children with PNS and has a close relationship with PNS. Our data suggest that miR-151-3o mav serve as a potential therapeutic target of PNS.relationship with PNS. Our data suggest that miR-151-3p may serve as a potential therapeutic tardet of RNS.
