有利于桑黄产胞内黄酮的液体发酵培养基配方优化 被引量:4

Optimization of Liquid Fermentation Medium Favorable for Production of Intracellular Flavone from Phellinus igniarius

摘要 黄酮是药用真菌桑黄的主要活性成分之一。研究桑黄液体发酵培养基中不同碳源、氮源、无机盐对桑黄菌丝体生物量及胞内总黄酮产量的影响,筛选出最佳碳源、氮源、无机盐种类,并在单因素试验的基础上利用Box-Benhnken设计和响应面分析法优化其用量,得到有利于桑黄产胞内黄酮的液体发酵培养基最佳配方为:葡萄糖29.6 g/L,黄豆粉10.2 g/L,硫酸镁(MgSO4·7H2O)2.3 g/L,酵母膏1.1 g/L,KH2PO41.0 g/L。使用该优化液体发酵培养基培养桑黄,其胞内总黄酮产量可达186.75 mg/L。建立的桑黄菌丝体胞内总黄酮产量与培养基4种组分浓度的回归模型拟合性好,可用于预测胞内总黄酮产量。 Flavone is the main active ingredient of medicinal fungus Phellinus igniarius. In this study, the effect of different carbon sources, nitrogen sources, and inorganic salts in liquid fermentation medium on mycelium biomass and intra- cellular flavone yield of P. igniarius was investigated to determine the optimal carbon source, nitrogen source and inor- ganic salt. Based on single-factor tests, Box-Benhnken design combined with response surface analysis was conducted to optimize the dosage of carbon source, nitrogen source and inorganic salt. The optimized medium formulation of liquid fermentation medium for intracellular flavone was as follows, glucose 29.6 g/L, soybean flour 10.2 g/L, MgSQ4.7H2Q 2.3 g/L, yeast extract 1.1 g/L, and KH2PQ4 1.0 g/L. The yield of intracellular flavone of P. igniarius cultured in the optimized medium reached 186.75 mg/L. The established regression models between the yield of intracellular flavone and the concentration of 4 ingredients in the medium had good fitting performance, and could be used to predict the yield of intracellular flavone from P. igniarius.
