摘要
目的采用Lowry法测定马铃薯中蛋白质提取物的含量。方法以国家蛋白质标准品为含量测定对照品,样品溶液和标准品溶液分别取1 ml,加碱性铜溶液5 ml,室温放置10 min,快速加入酚试剂0.5 ml,室温放置30 min,在波长650 nm处检测吸光度,外标法测定蛋白含量。结果国家蛋白标准品线性范围0.1~0.5 mg.ml-1,回归方程为Y=1.874 X+0.064(R=0.999),平均加样回收率为100.24%,RSD为1.63%。结论用该Lowry法测定马铃薯蛋白质提取物的含量,方法准确、简便、重复性好。
Objective To determine the protein content in potato extract by lowry method.Methods Using the national standard substance for protein content determination as the reference substance,5 ml copper ions in alkaline solution was added to 1 ml tested solution and mixed.After being placed at room temperature for 10 min,the mixture was added 0.5 ml phenol solution,placed at room temperature for 30 min and detected absorbance at the wavelength of 650 nm.The protein content was analyzed by the external standard method.Results The concentration of the standard solution was within the range of 0.1-0.5 mg · ml-1.A good liner relationship was observed.The regression equation was Y=1.874X+0.064(R=0.999),and the average recovery was 100.24%,with an RSD of 1.63%.Conclusion The improved method is accurate,simple and repeatable.
