摘要
建立了一种基于发酵液pH反馈控制γ-聚谷氨酸发酵中葡萄糖补加的方法,即:发酵过程中pH再次回升至7.0时,补加浓缩葡萄糖使发酵液中残糖达到1.5 g/L时停止补加。结果表明,利用该补料方法可较理想地将发酵液中葡萄糖浓度控制在1.0 g/L,发酵终产量达到42.5g/L,产率提高了46.8%,谷氨酸转化率提高了17.7%。
A pH feed-back-controlled fed-batch culture method was proposed for the production of γ-polyglutamic acid. When the pH was ascended to 7.0 again, substrate feeding was started ; and when the glucose concentration reached to 1. 5 g/L, the feeding was stopped. The result indicated that, the glucose concentration in the fermentation broth was maintained at 1.0 g/L, ideally. At the end of fermentation, 42.5 g/L γ-polyglutamic acid was produced. The γ-PGA productivity and glutamate conversion rate increased by 46.8% and 17.7% , respectively.
