摘要
分别以5、10、15、20、25Gy电子束剂量辐照百合品种Starfighter和Tiber的鳞茎,发现该剂量梯度明显抑制百合当代的营养生长和生殖生长,但不影响新鳞茎定植后第2代正常营养生长和开花,而且在C-15Gy第2代中发现花朵颜色变深、花瓣斑点长突起变短的变异株。以百合幼嫩叶片作为材料,应用RAPD技术分析不同剂量电子束辐照的扩增结果。11个试验组中,有5个随机引物扩增出清晰谱带282条,其中,32条为多态性谱带,多态率为11.35%。RAPD结果显示,电子束辐照百合鳞茎能够引起百合DNA变异,并且辐照剂量与诱变频率呈正相关关系,此外诱变频率还与试验品种、辐照部位及所处的生长期有关。
The bulbs of Lily Starfighter and Tiber were irradiated by electron beam in the dose of 5 Gy,10 Gy,15 Gy,20 Gy and 25 Gy,the results showed that Lily's contemporary vegetative growth and reproductive growth were severely inhibited,but the vegetative growth and blooming were unaffected in the second generation.Moreover,a variant plant with deeper flower color and shorter spots prominence on tepal was discovered in the second generation of C-15 Gy.Tender leaves of different irradiation dose were used as experimental material in RAPD analysis,5 random primers amplified 282 clear bands,32 of which were polymorphic bands,so the polymorphic rate of 11 experimental groups was 11.35%.The results of RAPD analysis indicated that electron beam irradiation on lily bulbs could induce variation,and the mutation rate was positively correlated to the irradiation dose,in addition,mutation rate was also related to experiment varieties,irradiate position and growing season.
