摘要
为探讨电子束辐照对睡莲"科罗拉多"的诱变效应,对其植株进行了剂量为5 Gy、10 Gy、15 Gy、20 Gy、25Gy、30 Gy、50 Gy的辐照,剂量率为1 Gy/s。将辐照后的植株与对照组在相同条件下种植,连续3年观察其营养生长和花部变化;并对不同辐照剂量的睡莲叶和花瓣DNA进行RAPD分析。结果显示,当年(2011年)各辐照组的营养体均存活,营养生长状况与对照组基本一致,但生殖生长上花量有所减少。次年(2012年)在10 Gy与15Gy组中各有1株的1朵花的花色有变化。第三年(2013年)10Gy组的同一变异株中出现1朵花保持了上一年的花色变异,花瓣大致为一半粉色一半黄色。与阴性对照比,RAPD分析10Gy剂量诱变的变异株,显示叶片RADP谱带发生或消失或新出现的明显变化。结果表明,电子束辐照可剂量依赖性地引起睡莲花量减少、花色改变,并诱发DNA产生变异。研究结果对电子束辐照培育睡莲优良品种提供了一定的依据。
Mutagenic effects of electron beam on plant growth,flowers,as well as DNA in water lily( Nymphaea sp. ‘Colorado') were investigated. Mature plants were exposed a beam of electrons at a dose of 5,10,15,20,25,30 and 50 Gy with the dose rate of 1 Gy / s,respectively. The irradiated plants and the control group were cultivated in the uniform conditions.The vegetative growth and the changes of floral organs were observed in 3 successive years. Alterations of DNA in leaves and flowers were detected using randomly amplified polymorphic DNA( RAPD) technique. The results showed that all the irradiated plants were alive and the growth of vegetative mass had no significant change in the same year( 2011). However,the number of flowers in all the irradiated groups was decreased. In each of the 10 Gy and 15 Gy groups,one plant emerged a flower with variant color respectively in the next year( 2012). Till to the third year( 2013),the same 10 Gy-variant plant still had a flower remained the variation feature,half of its petals in pink,and the others becoming yellow. RAPD polymorphisms became evident as disappearance or appearance of RAPD bands in the leaves and petals of the 10 Gy irradiated variant plant. The results showed that electron beam irradiation on water lily caused the decreased number,changed color of flowers and induced DNA variation,in a dose-dependent meaner. This research provides a basis for the breeding of water lily by electron beam irradiation.
