摘要
目的探讨肠三叶因子(ITF)对肠组织NF-κB及TNF-α 的调节及与肠损伤保护作用的关系。方法24只10日龄的Wistar幼鼠随机分为正常对照组(NS组),脂多糖(LPS)组,ITF组,每组8只。NS组予9g/L盐水1mL/kg腹腔注射;LPS组予LPS(5g/L)5mg/kg腹腔注射;ITF组予重组nT(5g/L)0.1mL/只+LPS5mg/kg腹腔注射。于腹腔注射后3h处死幼鼠,留取远端回肠组织,HE染色,光镜下行肠组织病理学检查。RT-PCR检测肠组织NF-κBmRNA表达水平。免疫组织化学检测肠组织NF-κB蛋白定位表达。ELISA法检测肠组织TNF-α 分泌水平。结果光镜下NS组肠组织结构正常,ITF组和LPS组均可见肠黏膜间质和上皮细胞水肿,ITF组较LPS组明显减轻;ITF组肠组织NF-κBmRNA和NF-κB蛋白表达均较LPS组明显下降(P均〈0.01);ITF组肠组织TNF-α 分泌较LPS组明显减少(P〈0.01)。结论ITF减轻肠组织损伤的保护作用可能与其下调NF-κBmRNA和蛋白的表达及降低炎性介质TNF-α 的分泌相关。
Objective To investigate the relationship of intestinal trefoil factor(ITF) for regulating of nuclear factor(NF) -κB and tumor necrosis factor(TNF) -α and the protective effect of intestinal injury. Methods Twenty-four 10-day Wistar rats were divided into 3 groups:the control group( NS group, n = 8), given 9 g/L sodium injection intraperitoneaUy, 1 mL/kg; the lipopolysaceharide (LPS) group ( n = 8 ), given-LPS ( 5 g/L), 5 mg/kg intraperitoreally; ITF group(n =8) ,given ITF(5 g/L) 0.1 mL/each plus LPS 5 mg/kg intraperitoreally. Rats were sacrificed 3 hours after injection. A segment of distal ileum was dissected. The pathologic changes of small intestine were observed under the optical microscope( hematoxylin-eosin staining). The expressions of NF-κB mRNA and protein were detected by reverse transcription polymerase chain reaction( RT-PCR), immunohistochemistry, respectively. TNF-α level in intestinal tissues was measured by enzyme linked immunoserbent assay. Results The structure of small imestine in the control group remained normal. The inflammatory cells infiltration and the edema of interstitial substance and epithelium were observed in LPS group and ITF group,while the change in the ITF group was significantly lower than that in LPS group. The expressions of NF-κB mRNA and protein in ITF group were significantly lower than those in LPS group( all P 〈 0. 01 ). The secretion of TNF-α in the ITF group was significantly lower than that in LPS group ( P 〈 0. 01l ). Conclusion Protective effects of ITF on intestinal injury are related to the down regulation of NF-κB mRNA and protein expressions and the reduction of the secrete of mediators of inflammation TNF-α.
