半枝莲和白花蛇舌草总多糖对S_(180)荷瘤小鼠的抗肿瘤作用研究 被引量:44

摘要 目的:探讨半枝莲和白花蛇舌草总多糖对S180荷瘤小鼠的抗肿瘤及其免疫调节作用。方法:采用MTT比色法检测肿瘤细胞的生长抑制率;流式细胞术分析细胞周期的改变;建立S180荷瘤小鼠模型,观察小鼠肿瘤的生长情况,计算抑瘤率、胸腺与脾脏指数;观察对荷瘤小鼠体液免疫功能及网状内皮系统吞噬功能的影响。以上动物试验在造模同时灌胃给予不同剂量的半枝莲和白花蛇舌草总多糖和环磷酰胺,模型组和正常组动物给予等容量的生理盐水。结果:浓度为25μg/mL、50μg/mL、100μg/mL、200μg/mL和400μg/mL的半枝莲和白花蛇舌草总多糖均可抑制S180肿瘤细胞株的体外增殖,且抑制作用呈浓度-效应依赖性,100μg/mL、200μg/mL和400μg/mL使G0/G1期细胞百分数增加,G2/M期细胞百分数下降;200mg/kg对S180荷瘤小鼠瘤重生长具有明显抑制作用,并且增加其胸腺指数和脾脏指数;200mg/kg和100mg/kg明显提高S180荷瘤小鼠的血清半数溶血素值,200mg/kg显著提高荷瘤小鼠的吞噬指数K及吞噬系数α值。结论:半枝莲和白花蛇舌草总多糖对S180荷瘤小鼠具有较强抗肿瘤作用,并可以提高其免疫系统功能。 Objective: To investigate the anti-tumor and immunomodulatory effects of polysaccharides from Herba Scutellariae Barbatae and Herba Hedyotis Diffusae on S180 tumor-bearing mice. Methods: Except for the normal group and model group, mice in polysaccharides groups were daily intragastrically administered with polysaccharides, and mice in cyclophosphamide group were daily intragastrically administered with cyclophosphamide. The tumor cell growth inhibition rate was measured by MTT colorimetry. Flow cytometry was used to analyze the cell cycle of S180 cells. The S180 xenograft tumor models were established for the observation of tumor growth inhibitory rate, spleen index and thymus index. Effects of the polysaccharides on the humoral immune function and the phagocytosis function of endothelial system of S180 tumor-bearing mice were assessed. Results: Polysaccharides(100, 200, 400μg/mL) could remarkably inhibit the proliferation of the S180 tumor cells in concentration-dependent manner. They also increased the cell count at G0/G1 phase and decreased the count at G2/M phase. Compared with the model group, polysaccharides(200mg/kg) could remarkably inhibit the tumor growth inhibitory rate and increase the spleen index as well as thymus index in mice. It could also elevate the clearance exponent K and phago- cytosis quotient a of S180 tumor-bearing mice. Polysaccharides(100, 200mg/kg) could increase the serum half hemolytic val- ue of S180 tumor-bearing mice. Conclusion: Polysaccharides from Herba Scutellariae Barbatae and Herba Hedyotis Diffusae have strong anti-tumor effects and could promote the immune function of the S180 tumor-bearing mice.
