摘要
目的:利用综合加权评分-Box-Behnken响应面法优选半枝莲的最佳提取工艺。方法:以甲醇浓度、液料比、提取时间为考察因素,利用Box-Behnken响应面法建立三因素三水平并设计试验,通过综合加权评分法对半枝莲中的化学成分野黄芩苷、黄芩苷、芹菜素、木犀草素、原儿茶酸、对香豆酸、肉桂酸分别赋予其权重,并以综合加权评分值为评价指标,考察三因素对半枝莲提取效果的影响,优选出最佳提取工艺,并对该工艺进行验证。结果:最佳提取工艺参数为:甲醇浓度73.19%、液料比19.12(mL/g)、提取时间1.99 h,在此条件下综合加权评分值为95.30。结论:优选出的提取工艺稳定可靠,建立的提取方法简单可行,对半枝莲提取工艺的方法研究具有参考价值。
Objective:To optimize the extraction process of Scutellaria barbata by comprehensive weighted score combined with Box-Behnken response surface methodology.Methods:Box-Behnken response surface methodology was applied to design a three factors and three levels experiment with methanol concentration,liquid-solid ratio and extraction time as investigation factors.The weight of scutellarin,baicalin,apigenin,luteolin,protocatechuic acid,p-coumaric acid and cinnamic acid of Scutellaria barbata were given by comprehensive weighted scoring method,which were taken as evaluation parameters to investigate the influence of three factors on the extraction effect of Scutellaria barbata,the best extraction conditions were selected and verified.Results:The optimum extraction conditions were 73.19%for methanol concentration,19.12(mL/g)for liquid-solid ratio and 1.99 h for extraction time,the comprehensive weighted score was 95.30 under the above conditions.Conclusion:The optimized extraction process is stable and reliable,the proposed extraction method is simple and feasible,which provides reference basis for extraction process study of Scutellaria barbata.
