复合诱变抗性筛选法选育高转化甘油产1,3-二羟基丙酮菌株

Screening of Mutant Strains for Converting Glycerol to Dihydroxyacetone by UV and ^(60)Co Irradiation Combining Drug-Resistance Technique

摘要 在测定不动杆菌(Acinetobacter sp.)Asp16菌株链霉素最低杀菌浓度的基础上,对菌株进行UV和60 Co诱变处理后,利用链霉素抗性平板筛选获得1株遗传性能稳定、高转化甘油产1,3-二羟基丙酮的突变株Asr168,将突变株在5.0g·L-1酵母膏、140.0g·L-1甘油、7.0g·L-1 CaCO3组成的发酵培养基中于30℃、250r·min-1摇床培养50h,1,3-二羟基丙酮的产量达到136g·L-1,比出发菌株提高200%。表明通过UV和60 Co诱变,结合链霉素抗性平板筛选,可大幅提高不动杆菌转化甘油产1,3-二羟基丙酮的能力。 Ni-Ce-B Amorphous alloy was prepared at 50 *C by the reduction of NiCl2 6H20 and Ce(NO3)3 6HzO using KBH4 as reduction agent in OP-lO-SDS/n-butanol/cyclohexane/water microemulsion, and its morphology,structure, thermal stability and crystallization properties were studied by TEM, SEM, TG-DSC and XRD. The effect of size of aqueous nucleus on average particle size was investigated through changing the composition of aqueous phase. The results showed that Ni-Ce-B amorphous alloy with average particle size of 30 nm could be prepared,and the particle size distribution was uniform with good monodispersity. The average particle size increased with w value and Rw value increasing. The crystallization temperature of Ni-Ce-B amor- phous alloy was significantly higher than that of Ni-B amorphous alloy by 360 ℃.
