摘要
建立胃泰咀嚼片中黄芪甲苷的质量比测定方法.采用高效液相色谱-蒸发光检测器定量分析黄芪甲苷的质量比,使用Alltima-C18(5μm,4.6 mm×250 mm)色谱柱,流动相为甲醇-水(68∶32),流速1mL/min;蒸发光散射检测器(ELSD)飘移管温度50℃,载气压力1.58 Bar,放大系数(Gain)为10;进样量为20μL.表明黄芪甲苷在0.95～22.8μg之间呈良好的线性关系,r=0.999 00;加样回收率为98.97%,RSD为0.64%,在常温下8 h内稳定性较好.本方法简便快速,准确可靠,可作为胃泰咀嚼片的质量控制方法.
To establish an method for the determination of astragaloside in Weitai chewable tablets. Conducted determination of astragaloside in this drug by HPLC - ELSD. Chromatogram condition ： selected Alhima - Cl8 （5μm, 4.6 mm ×250 mm） ; mobile phase ： methanol -water（68：32）, speed of flow was 1.0 mL/min;the ELSD conditions were as follows： pressure of compressed air 1.58 bar, drift temperature 50℃ and gain 10. The injection volume was 20 μL. The linear range of astragaloside was 0.95- 22.8 μg （ r = 0. 999 0） and the average recovery rate was 98.97% ,RSD =0.64%, this substance in eight hours at room temperature stability was good. This method was accurate, reproducible and stable, and could be used for quality control of Weitai chewable tablets.
