摘要
目的探讨体外循环(CPB)对大鼠血脑屏障(BBB)功能及脑血管内皮紧密连接蛋白表达的影响。方法 24只成年雄性SD大鼠,随机分为假手术组(S组)和体外循环组(C组)。S组进行相应的麻醉和手术操作但不进行CPB,C组进行CPB 60 min并继续观察2 h。每组随机取6只测脑组织伊文思蓝(EB)含量,另6只做脑组织含水量测定、紧密连接蛋白Oc-cludin和ZO-1的表达以及大鼠BBB超微结构的观察。结果与S组比较,C组大鼠海马区脑组织EB含量和含水量升高,差异均有统计学意义(P<0.05)。与S组比较,C组大鼠脑血管内皮紧密连接蛋白Occludin和ZO-1表达水平下降,差异均有统计学意义(P<0.05)。S组BBB基膜均匀一致,完整性良好,内皮细胞间的紧密连接完整;C组基底膜变厚,微血管腔狭窄,内皮细胞间紧密连接不清楚。结论 CPB使大鼠BBB的结构发生变化、通透性增加,其机制可能与大鼠脑血管内皮紧密连接蛋白改变有关。
OBJECTIVE To investigate the effects of cardiopulmonary bypass(CPB) on blood-brain barrier(BBB) and the expression of tight junction(TJ) protein of cerebral vascular endothelium in rat model.METHODS 24 adult male Sprague-Dawley rats were randomly divided into two groups: group S served as shame-operated group without CPB;the rats of group C underwent 60 minutes of CPB and were observed 2 hours later.The Evans blue content of brain tissue was measured in 6 rats of each group,the water content of brain tissue,the Occludin and ZO-1 expression of brain tissue,and the ultrastructural of BBB were observed in the another 6 ones of each group.RESULTS Compared with group S,the Evans blue content and water content of group C significantly increased(P0.05),the expression of Occludin and ZO-1 of group C significantly decreased(P0.05).The basement membrane of BBB of group S was uniform,but that of group C was swollen.The TJ between endothelia of group S was intact,but that of group C was unclear.CONCLUSION CPB destroys the structure of BBB and increases the permeability of it,which may have relationship with the change of TJ protein.
