摘要
目的探讨枣花丹合剂醇提物(CZHD)对小鼠镇静、催眠作用的影响。方法本实验将50只小鼠随机分为生理盐水组、安定组、CZHD高、中、低剂量组5组,每组10只;采用醇提及微波辅助法的方法来充分提取其有效成分,通过测定小鼠的自主活动次数,小鼠睡眠时间及对抗3-MP诱导惊厥程度等指标来观察CZHD对正常小鼠的镇静催眠作用。结果 CZHD高[40g/(kg.d)]、中[20g/(kg.d)]、低[10g/(kg.d)]剂量组,均能增加入睡小鼠的动物数(P<0.05);与生理盐水组相比,CZHD高、低剂量组能相对降低小鼠自主活动次数(P<0.05),延长戊巴比妥钠阈剂量睡眠时间(P<0.05),明显降低3-MP所诱发的小鼠惊厥率;但与与安定组相比,作用无统计学意义(P>0.05)。结论枣花丹合剂醇提物对小鼠具有明显的镇静催眠效果,其中高剂量组及低剂量组具有明显抑制其自主活动及对抗惊厥的作用。
Objective To explore the Zao Huadan mixture of alcohol extract （CZHD） has sedative and hypnotic effects in mice. Methods 50 mice were divided into five groups ： normal saline group ; diazepam group ; High dose group ; In the dose group and Low dose group of the CZHD. Each group was 10 mice. Prescription drugs under the different solubility of the active ingredient were mentioned and the method of alcohol dissolved by microwave assisted was used to fully extract the active ingredients. We recorded the number of moving of the mice with spontaneous activity instrument, the mice sleeping time was measured by turning over reflecting method and the sedative effect was observed by the anti - convulsion test that was induced by 3 - MP. Results CZHD high （40g/d）, in （ 20 g/ d） ） ,low （ 10 g/d） dose groups could increase the number of animals to sleep in mice （ P 〈 0.05 ）. Compared with the saline group, CZHD high and low dose group relative decrease spontaneous activity in mice （ P 〈 0. 05 ） , extend the threshold dose of sodium pentobarbital sleeping time （ P 〈 0. 05 ） and significantly lower 3 - MP induced seizure rate of mice （ P 〈 0.05 ） , but they had no significant effect （ P 〉 0. 05 ） compared with the diazepam group. Conclusion TBZWZao Huadan mixture of alcohol extract has significant sedative and hypnotic effects on mice. High dose group and low dose group decrease its spontaneous activity and inhibit the 3 - MP induced seizure.
