摘要
目的对红豆杉多糖进行分离、纯化并对其抗肿瘤活性进行测试。方法使用热水浸提分级醇沉的方法提取多糖,收集90%醇沉多糖,使用Sevag法除蛋白质,对得到的粗多糖使用DE-52离子交换纤维素和丙烯葡聚糖凝胶Sephacryl S-300进行色谱分离,对分离的精多糖使用凝胶排阻色谱法测定相对分子质量(Mr),红外光谱检测糖构型,MTT法测定其体外抗肿瘤活性。结果分离得到两个精多糖TMP90W和TMP90S,Mr分别为11 700和18 500。初步活性测试结果表明,TMP90W对人乳腺癌细胞MCF-7和人肺癌细胞A549均具有一定的抑制作用。结论 TMP90W具有作为抗肿瘤或辅助抗肿瘤候选药物的潜力。
Purpose To isolate and identify the polysaccharides of Taxus chinensis and to determine their antitumor activities in vitro.Methods Using hot water extraction and alcohol precipitation to get the crude polysaccharides,removing protein by Sevag method,and further purifying by column chromatography packed with DE-52 and Sephacryl S-300 HR.The homogeneity and molecular weights were evaluated and antitumor activities in vitro were determined by MTT.Results TMP90W with molecular weights of 11.7 kDa could restrain the growth of the tumor cells.Conclusion The polysaccharides isolated from T.chinensis could be a potential antitumor agent.
