摘要
采用SDS抽提法从酵母中提取蔗糖酶,通过正交试验对SDS摩尔浓度、pH值、提取温度、提取时间进行优化,确定最佳提取工艺条件是SDS摩尔浓度为0.5mmol/L,温度40℃,提取时间10h,pH值5.5。制得的粗酶经醇沉后,采用Q-琼脂糖凝胶FF阴离子交换层析方法得到纯化的酵母蔗糖酶,纯化倍数为18.7倍,对蔗糖酶的性质也进行了研究。
The experiment extracted invertase from brewer's yeast using SDS extraction.Through the orthogonal experiment on the SDS concentration,pH value,extraction temperature,extraction time and other conditions,it optimized the extraction process,determine the optimum extraction conditions were:SDS concentration 0.5mmol/L,temperature for 40℃,time for 10h,pH value for 5.5.The crude enzyme obtained was precipitated by ethanol,and used Q-SepHarose FF anion-exchange column chromatography method of increasing ionic strength to get purificated yeast invertase,purification multiple was of 18.7 times and research its quality.
