摘要
采用复合蛋白酶(胰蛋白酶、碱性蛋白酶)对大麦虫蛋白进行控制酶解,研究酶解时间、酶活比、底物浓度、酶解温度、pH与加酶量等对酶解反应的影响;采用中心组合试验设计,优化酶解工艺;并对大麦虫酸性多肽清除自由基能力进行了测定评价。结果表明,酶解最佳条件为底物浓度5%,时间120 min,胰蛋白酶∶碱性蛋白酶1∶1,pH 8.71,温度48℃,加酶量16.91 mg/g。该酸性多肽对(O2-.)、DPPH.自由基有很好的清除作用,具有很强还原力。利用优化工艺制得酸性肽得率为79.05%。
In order to obtain the bioactive peptides,the barley pest protein was hydrolyzed and the antioxidant of its hydrolyzed liquid were determined.The barley pest protein was hydrolyzed by the mixture of trypsin and alkaline protease,substrate concentration,time,the effects of ratio of protease energy,temperature,pH and dosage of protease on enzymatic reaction were studied,the conditions of enzymatic reaction were optimized by central composite experimental design.Then antioxidative activity of the hydrolyzed liquid were determined by its scavenging activity and reducing activity.The optimum hydrolyzed conditions were: substrate concentration 5%,time 120 min,ratio of protease energy 1∶1,pH 8.71,temperature 48℃and dosage of protease 16.91 mg/g.The hydrolyzed liquid has strong scavenging activity on(O2^-·)and DPPH·,and strong reducing activity.Under these conditions,YASP was 79.05%.
