摘要
对沉淀剂的体积比、沉淀剂与溶血液的体积比和沉淀时间对羊血红蛋白去除效果的影响进行了研究,确定了最优工艺参数.采用中心试验组合设计和响应面分析法,得出的最优组合为A2B2C2,即沉淀剂为95%的乙醇与丙酮、沉淀剂与溶血液,体积比均为1∶1,沉淀时间为30 min.除血红蛋白是羊血超氧化物歧化酶(Superoxide Dismutase,SOD)提取的关键,用响应面法工艺除羊血红蛋白后得到的SOD粗酶液,活性为302.59 U/mg.
After the study on the effect of the ratio of precipitant, the ratio of precipitation agent and volume of blood and settling time on removing hemoglobin and extraction of SOD, its optimal technique parameter is defined. Box-Benhnken central combination design and response surface methodology analysis of the SOD activity are used, and the optimal combination of orthogonal experiment is A2 B2 C2, that is, the ratio of precipitation agent and volume of blood is 1 1.95% ethanol and acetone volume ratio is 1 : 1, the extraction time is 30min, and activity of SOD is 302.59 U/mg Protein.
