摘要
研究不同链长花生蛋白肽的体外抗氧化活性。采用碱性蛋白酶Alcalase水解花生蛋白质,经超滤获得分子量介于5000~3000Da之间以及分子量小于3000Da的2种花生蛋白肽混合物PPH-1和PPH-2;以VE为对照,分别测定肽对脂质过氧化的抑制性能、还原能力和对二苯代苦味酰基自由基(DPPH·)清除能力。在抑制脂质过氧化方面,浓度为0.05mg/mL时,PPH-1的活性低于VE,PPH-2的活性则高于VE,在第7天,PPH-1和PPH-2对脂质过氧化的抑制率分别为69.1%和84.5%;在还原能力方面,PPH-2的活性大于PPH-1,但二者均低于VE,在浓度为1mg/mL时,PPH-1和PPH-2的还原能力分别与浓度为0.013,0.021mg/mL的VE相当;在自由基清除能力方面,随着花生蛋白肽浓度的增加,其对二苯代苦味酰基自由基清除能力增大,在浓度达到50mg/mL时,PPH-1和PPH-2对二苯代苦味酰基自由基的清除率分别是35.2%和60.3%。PPH-2的分子量主要分布在2大区域,其氨基酸含量达到91.15%。分子量小于3000Da的花生蛋白肽具有较好的抗氧化活性。
To investigate the antioxidative activity in vitro of peanut peptide with different chain length. The peanut protein was hydrolyzed by Alcalase. Hydrolysate was ultrafiltered and two kinds of peptide (PPH-1 and PPH 2) were obtained, whose molecular weight distribution were between 5 000 and 3 000 Da and lower than 3000 Da. Compared with those of VE, the inhibitory effect on the auto-oxidation of linoleic acid, reducing power and DPPH · scaven ging activity of the peptides were measured. In the inhibitory effect on the auto-oxidation of linoleic acid, PPH-1 was lower than VE, while PPH-2 was superior to VF. At the seventh day, the inhibitory rates of PPH 1 and PPH 2 with the concentration of 0.05 mg/mL were 69. 1% and 84. 5%, respectively. In reducing power, PPH-1 was superior to PPH 2, but lower than VE. At the concentration of 1 mg/mL, the reducing powers of PPH-1 and PPH-2 were corresponded to VE whose concentrations was 0.018 mg/mL and 0.03 mg/mL respectively. In the DPPH· scavenging activity, as the concentration increased, the scavenging activity increased. At 50 mg/mL, the DPPH· scavenging activity of the PPH-1 and PPH- 2 were 35. 2% and 60. 3% The molecular weight distribution of PPH-2 contained two main region and the amino acid of the PPH-2 were 91.15%. PPH-2 with molecular weight lower than 3 000 Da had better antioxidative activity.
