罗红霉素与碘在乙醇中的荷移反应及其测定 被引量:2

摘要 目的建立测定制剂中罗红霉素含量的新方法。方法用分光光度法研究了罗红霉素与碘在乙醇中的荷移反应。结果荷移反应形成的络合物在290和360nm处有2个吸收峰,表观摩尔吸光系数分别是1.553×10^3和1.104×10^3L/(mol·cm),药物浓度在2~500mg/L范围内服从比耳定律,用摩尔连续变化法和摩尔比法测得荷移络合物中罗红霉素与碘分子的摩尔比是1:3,罗红霉素浓度为250mg/L时,相对标准偏差(RSD)为0.907%(n=9)。用所研究方法测定了罗红霉素胶囊和分散片的含量,回收率分别为98.86%,97.12%。结论方法灵敏,快速,准确,与药典方法比较结果一致。 Objective To establish a method for rapid determination of the content of roxithromycin in preparation. Methods The charge transfer reaction between roxithromycin and iodine in alcohol medium was investigated by spectrophotometry. Results The apparent molar absorptivity of complex was 1. 553×10^3 L/( mol·cm) at 290nm 1. 104 × 10^3 L/(molocm) at 360 nm. Beer's law was obeyed in the concentration range of 2-500 mg/L. The composition ratio and stability constant of charge transfer complex were 1 : 3 respectively. The average standard deviation of nine determinations was 0. 907% when the concentration of roxithromycin was 250 mg/L. The method has been successfully applied to the determination of roxithromycin dispersible capsules and dispersible tablets. The average recovery was over 97%. Conclusion The proposed method is accurate, sensitive and rapid, the results are in good agreement with those obtained with the official method.
