DNA聚合酶β与遗传不稳定性研究现状 被引量:2

摘要 DNA聚合酶β(polβ)是碱基切除修复系统(BER)中的核心成分,在DNA损伤修复及维持基因组稳定性和完整性中起着重要的作用。由于其缺乏3′~5′的校读功能,polβ在DNA合成中复制保真度较低,有关polβ在遗传不稳定性中的作用规律及其在肿瘤发生中的作用机制研究结果不尽一致。本文对polβ的结构与功能、polβ与基因组遗传不稳定性,以及polβ在肿瘤细胞组织中异常表达和突变的研究现状进行概述。 DNA polymerase β (pol β), a predominant enzyme involved in DNA base excision repair (BER), plays an important role in repair of DNA damage and maintaining stability and integrality of genome. Pol β lacks 3' to 5' proofreading activity, which contributes to its low fidelity in DNA synthesis. There are different results about action rule of pol β in genetic instability and mechanism of pol β in tumorigenesis. The structure and function of pol β the relationship between pol β and genome instability, and abnormal expression and mutation of pol β in tumor were reviewed.
