川芎嗪对大鼠颈总动脉损伤后血管狭窄的干预作用 被引量:1

摘要 目的观察川芎嗪(TMP)对血管损伤后血浆血管紧张素Ⅱ(AngⅡ)、血管血小板源生长因子(PDGF)与细胞外信号调节激酶(ERK1/2)表达的影响。方法制备大鼠血管再狭窄模型,观察颈总动脉形态学变化,放射免疫法测定血浆AngⅡ含量,免疫组化法观察血管PDGF-B、ERK1/2表达。结果假手术组动脉各层结构正常,颈总动脉损伤大鼠VSMC大量增殖,内膜显著增厚,TMP处理后,VSMC增殖程度降低;与假手术组比较,手术组血浆AngⅡ含量显著升高(P<0.01),PDGF-B、ERK1/2表达显著增强(分别P<0.01)。TMP明显降低AngⅡ含量、PDGF-B与ERK1/2的表达(P<0.05)。结论TMP通过降低血浆AngⅡ水平、抑制PDGF-B与ERK1/2表达实现抗血管再狭窄的作用。 [Objective] To observe the effects of Tetramethylpyrazine (TMP) on angoitensin Ⅱ (AngⅡ) level and vessel platelet-derived growth factor-B (PDGF-B) and extracellular signal regulated kinase (ERK1/2) expression in carotid artery injury rats. [Methods] Restenosis model was established. The morphologie change and AngⅡ level of plasma were measured. The expression of PDGF-B and ERK1/2 was detected by immunohistoehemieal technique. [Results] Compared with sham group, the sections of injured carotid artery showed significantly intimal thickening with large numbers of vascular smooth tousle cell (VSMC) proliferating throughout intima and media, the level of AnglI was obviously increased (P 〈0.01), and the expression of PDGF-B and ERK1/2 was significantly increased [(P 〈0.01), respectively] after operation. VSMCs proliferating was inhibited and the AngII level, PDGF-B and ERK1/2 expression were decreased obviously by TMP. [Conclusion] Maybe TMP is participate in the therapy of vascular restenosis through inhibiting AngII level, PDGF and ERK1/2 expression.
