Content Analysis of Polysaccharides and Nucleosides in Stroma of Cultured Cordyceps militaris and Culture Medium

摘要 目的建立分析北虫草中多糖和核苷类成分含量的方法,并对比分析北虫草子座和培养基中的成分。方法用硫酸-苯酚法测定北虫草中多糖含量;用RP-HPLC法同时测定北虫草中尿苷、腺苷和虫草素含量。结果北虫草子座中多糖含量分别为4.67%和4.34%,培养基中为3.42%。北虫草2006年样品与2007年样品比较,尿苷(0.267%vs0.063%)、腺苷(0.247%vs0.082%)和虫草素(0.054%vs0.166%)含量均有显著差异,2007年样品子座与培养基相比,尿苷(0.063%vs0.052%)和腺苷(0.082%vs0.117%)含量无显著差异,培养基中虫草素含量(0.318%)显著高于子座(0.166%)。结论建立了北虫草多糖和核苷类成分定量分析方法。该方法简便、可靠,可准确测定北虫草中多糖和3种核苷类成分的含量。 Objective To establish methods to determine the contents of polysaccharides and nucleosides in Cordyceps militaris and to compare the contents of them in the stroma and culture medium. Methods The contents of polysaccharides in Cordyceps militaris were determined by phenol-sulfuric acid method. The contents of uridine, adenosine and cordycepin were simultaneously determined by RP-HPLC method. Results The contents of polysaccharides in the stroma of Cordyceps militaris were 4.67 % and 4.34 %, while 3.42 % in the culture medium. Comparing the sample of Cordyceps militaris obtained in 2006 with that in 2007, there were large differences in the contents of uridine (0.267 % vs 0.063 %), adenosine (0.247 % vs 0.082 %) and cordycepin (0.054 % vs 0.166 %). In the stroma and culture medium of the sample obtained in 2007, the contents of uridine and adenosine were 0.063 % and 0.052 %, 0.082 % and 0.117 %, respectively, which showed no obvious differences. But the content of cordycepin in the culture medium was 0.318%, which was much higher than 0.166 % in the stroma. Conclusion The quantitative analysis methods can be obtained for the determination of polysaccharides and nucleosides in Cordyceps militaris. And these methods are convenient and reliable for the accurate determination of polysaccharides and three kinds of nucleosides in Cordyceps militaris.
