人参茎叶皂苷对体外条件下小鼠骨髓间充质干细胞和粒-巨噬系祖细胞增殖的影响 被引量:9

摘要 目的:探讨人参茎叶皂苷(GSL)在体外培养条件下对小鼠骨髓间充质干细胞(MSC)和粒-巨噬系祖细胞(CFU-GM)生长的影响。方法:在小鼠骨髓细胞液体培养体系和半固体培养体系中加入不同浓度的GSL,应用体外集落形成法检测GSL对小鼠MSC和CFU-GM增殖的影响。结果:在培养体系中GSL浓度为25μg/ml时,MSC和CFU-GM集落产率与对照组无明显差异(P>0.05);50~200μg/ml的范围内,MSC和CFU-GM集落产率显著高于对照组(P<0.05),其中浓度为100μg/ml时效应最明显;但在400μg/ml时,MSC和CFU-GM集落产率却低于对照组(P<0.05)。结论:GSL在一定浓度范围内对体外培养条件下的小鼠骨髓MSC具有生长促进作用,而在高浓度时表现出生长抑制作用。 Objective: To investigate the effects of ginsenoside of stem and leaf (GSL) on the proliferation of murine bone marrow mesenchymal stem cells (MSC) and colony - forming units for granulocytes and macrophages ( CFU - GM) in vitro. Methods. The colony - forming efficiencies of MSC and CFU - GM were evaluated in murine bone marrow cell cultures with or without the addition of GSL respectively. Results : There was not any significant difference in the colony yields of MSC and CFU - GM between the 25mg/ml GSL groups and controls. The colony - forming efficiencies were significantly increased in the MSC and CFU - GM cultures with 50mg/ ml to 200mg/ml GSL and the maximal effect was observed at 100mg/ml. However, the colony- forming efficiencies of MSC and CFU - GM at 400mg/ml of GSL were significantly lower than control groups. Conclusion: GSL over a range of concentrations has enhancing effects on the in vitro proliferation of murine bone marrow mesenchymal stem cells and colony - forming units for granulocytes and macrophages, whereas growth - inhibitory effects are found at higher concentrations.
