摘要
目的确定制备的平均相对分子量分别为8000D、5200D和4000D左右低分子硫酸皮肤素(LMWDS1、LMWDS2和LMWDS3)的结构。方法采用紫外(UV)、红外(IR)及核磁共振(13C-NMR、1H-NMR)图谱分析。结果LMWDS1还含有部分降解的-(1→3)-2-乙酰氨基-2-脱氧-4-O-硫酸-β-O-半乳糖;LMWDS2、LMWDS3中(1→4)-O-α-L-艾杜糖醛酸-(1→3)-2-乙酰氨基-2-脱氧-4-O-硫酸-β-O-半乳糖和(1→4)-2-O-硫酸-α-L-艾杜糖醛酸-(1→3)-2-乙酰氨基-2-脱氧-6-O-硫酸-β-O-半乳糖含量降低,而(1→4)-2-O-硫酸-α-L-艾杜糖醛酸-(1→3)-2-乙酰氨基-2-脱氧-4-O-硫酸-β-O-半乳糖的含量明显升高,从而导致了LM-WDS分子中硫酸根/羧酸根的比值升高。结论各LMWDS中均含有DS的主体双糖部分,随着降解程度的增加,双糖单位上某些取代基上的基团会发生变化。
Aim To identify the structures of low molecular weight Dermatan sulfate (LMWDSs). Methods According to the spectra of UV, IR, ^13 C-NMR and ^1H-NMR of all kinds of LMWDSs. Results The spectra proved the disaccharide unit structure of DS to be (1-4) -O-α-idopyranosyluronic acid- ( 1-3 ) -O-2-acetamido-2 - deoxy-4-O- sulfate-β-O-galattopyranosyl, as was the case with the natural one. The spectra also showed that with the decrease of molecular weight, the content of the above disacccharide unit in LMWDS decreased, but the content of (14) -2-O- sulfate-α dopyranosyluronic acid-( 1-3 )-O-2-acetamido-2-deoxy-4-O-sulfate-β-O-galattopyranosyl increased,pushing the ratio of SO3/C00. Conclusion Each of the LMWDSs took the disaccharide unit as the main part of the structure. As the molecular weight of LMWDSs decreased, the content of some disaccharide unit changed.
