创伤弧菌脓毒症病理组织学及血清细胞因子动态变化研究 被引量:6

摘要 目的探讨大鼠创伤弧菌(VV)脓毒症时病理组织学及血清细胞因子的动态变化情况。方法制作大鼠VV脓毒症模型(VV组),光镜下观察染菌大鼠主要脏器病理改变,ELISA法测定染菌后各时间点大鼠血清肿瘤坏死因子-α(TNF-α)和白细胞介素-10(IL-10)含量。结果染菌后16h光镜可见肺充血、出血明显,间质弥漫性炎细胞浸润,肺泡间隔形态改变,有局灶性坏死,肺纤维组织增生;肌肉呈严重蜂窝织炎,可见肌纤维断裂,有灶性坏死。染菌后2h起血清TNF-α含量明显升高(P<0.05);染菌后9h起血清IL-10含量升高(P<0.05)。结论VV脓毒症可致肺、肌肉等多组织脏器损害,VV脓毒症早期血清中以促炎因子TNF-α升高为主,脓毒症中后期时抗炎因子IL-10逐渐增多。 Objective To investigate the histopathology and changes of serum cytokines in rat with Vibrio vulnificus sepsis. Methods Vibrio vulnificus sepsis model (VV groups) was induced by injection of Vibrio vulnificus in SD rats. The histopathological changes were observed in light microscope, the levels of serum TNF-α and IL-10 were measured by enzyme linked immunosorbent assay (ELISA) at the different times. Results Under light microscope, congestion, haemorrhage and inflammatory cells infiltration were observed in lung; dermatocellulitis, focal necrosis and breakage in muscle fiber were observed in muscle. Compared with the control group, TNF-α levels were increased markedly after 2h and IL-10 levels were raised significantly after 9h (P〈0.05) in VV group (P〈0.05). Conclusion Our data suggest that VV sepsis could lead to damage in multiple organs, serum TNF-α levels increase significantly in the early period of VV sepsis and IL-10 levels raise later.
